An efficient strategy for the extraction and purification of lignans fromSchisandra chinensisby a combination of supercritical fluid extraction and high-speed counter-current chromatography

Abstract: An efficient strategy for extracting and separating five lignans from Schisandra chinensis (Turcz.) Baill has been developed using supercritical fluid extraction (SFE) and high-speed counter-current chromatography (HSCCC) in the present study. First, the extraction was performed by a preparative SFE system under 15 MPa of pressure at 36°C for 4 h. Then, the SFE extract was successfully separated and purified by HSCCC with a two-phase solvent system composed of n-hexane/ethyl acetate/methanol/water (6:4:5:5, 6:… Show more

“…Generally, strategies for the separation and purification of natural products have been based on chromatographic techniques such as column chromatography on silica gel, Rp‐18, and Sephadex LH‐20 . Recently, some strategies using preparative HPLC and high‐speed countercurrent chromatography have been reported. To obtain a sufficient amount of high‐purity compound for structural and pharmacological analyses, tedious procedures, including large amounts of extraction solvents, repeated cycles of extraction, fractionation, and purification steps, are necessary.…”

“…CCC is a support‐free liquid–liquid partition chromatography without the use of a solid support matrix . Recently, the method has been successfully applied to separate and analyze the various polyphenols from plant extracts []. However, there has been no report about the use of CCC for the isolation and purification of isomer of resveratrol dimer from wine grape.…”
