An E3-like Factor that Promotes SUMO Conjugation to the Yeast Septins

Abstract: Covalent attachment of the ubiquitin-related protein SUMO to other proteins participates in many processes including signal transduction, transcriptional regulation, and growth control. We report the characterization of Siz1 as an E3-like factor in the SUMO pathway. Siz1 is required for SUMO attachment to the S. cerevisiae septins in vivo and strongly stimulates septin sumoylation in vitro. Siz1 and the related protein Siz2 promote SUMO conjugation to different substrates at different stages of the cell cycle … Show more

“…Preventing SUMO attachment to septins Cdc3 and Cdc11 by mutagenesis of the relevant Lys residues to Ala causes 'old' septin rings to persist. By contrast, preventing SUMO attachment to the same septins by inactivation of the major SUMO ligase (by a siz1Δ mutation) does not cause the same phenotype [63]. These newer findings cast doubt on the suggestion that SUMOylation is required for disassembly of septin rings after completion of cytokinesis.…”

“…This defect was further exacerbated in siz1 siz2 slx5 or siz1 siz2 slx8 triple mutants (Table 2). On their own, siz1 siz2 double mutants displayed a synthetic growth defect [10,42] with a doubling time (DT) of 123 min, and loss of SLX5 or SLX8Δ in this background exacerbated this defect (DT = 200 min). The fact that all of these proteins contain RING-or SP-RING motifs suggested that Siz1 and/or Siz2 may functionally overlap with, or regulate the levels of Slx5 and Slx8.…”

“…The homologous Siz1/Ull1 and Siz2/Nfi1 proteins were the first SUMO E3 ligases to be identified [10]. These proteins share a RINGfinger related sequence motif, the SP-RING domain, with the PIAS1 (Protein Inhibitor of Activated Signal transducer and activator of transcription) protein of human cells [11,12].…”
