Background: Ganoderma fungus is rich in terpenoids. These compounds are known for their anti-hyperglycemic activities. However, the study of terpenoids as the secondary metabolite from Ganoderma as a dipeptidyl peptidase-4 (DPP-4) inhibitor remains unexplored. In addition, we examined the α-glucosidase inhibition activity. Objective: This study aimed to isolate the major terpenoid from non-laccate Ganoderma and examined its inhibitor activity on DPP-4 and α-glucosidase enzymes, and its interaction. Methods: The compound was isolated using column chromatography from Ganoderma australe. The structure of the isolated compound was confirmed by 1H and 13C nuclear magnetic resonance spectroscopy, while the inhibitory activity was evaluated using an enzymatic assay. The interaction of the isolated compound with DPP-4 and α-glucosidase enzymes was investigated using an in silico study. Results: The isolated compound was identified as stellasterol; IC50 values for DPP-4 and α-glucosidase inhibitor were 427.39 µM and 314.54 µM, respectively. This study revealed that the inhibitory effect of stellasterol on DPP-4 enzyme is through hydrophobic interaction, while the α-glucosidase enzyme is due to the interaction with six amino acids of the enzyme. Conclusion: Stellasterol is the major component of the steroid from G. australe. Enzyme inhibitory assay and in silico study suggest that stellasterol may contribute antidiabetic activity with a mechanism closer to acarbose rather than to sitagliptin.

Ganoderma sichuanense (Ganodermataceae) is a medicinal mushroom originally described from China and previously confused with G. lucidum. It has been widely used as traditional medicine in Asia since it has potential nutritional and therapeutic values. We collected 8 specimens of Ganoderma species from Thailand and show that they represent the first record of G. sichuanenese for Thailand. In this paper, we describe our specimens of Ganoderma sichuanense based on fresh basidiomes, and provide line drawings and photographs. The data from macro-and microscopic features are consistent with the characteristics of the species. Analysis of ITS sequence data indicates that the Thai collections cluster in same species clade as the epitype of G. sichuanense.

Mycelial microstructures of taxa of the genus Ganoderma from the IBK Culture Collection of Mushrooms at the M.G. Kholodny Institute of Botany NASU were investigated by light microscopy and scanning electron microscopy. In total, seven strains of the following five species were studied: G. сarnosum, G. resinaceum, G. sinense, G. tsugae, and G. oregonense. Mycelium of all these species consists of thin-walled septate hyphae of different size. Characteristic microstructures for vegetative mycelia were observed: clamp connections, swollen hyphae, mycelial pellicle, mycelial cords, staghorn (coralloid) hyphae, and chlamydospores. For G. carnosum the following mycelial microstructures were found: single clamp connections with or without anastomoses, double clamp connections with anastomoses and rarely swollen hyphae; for G. resinaceum and G. sinense-single clamp connections, large number of staghorn hyphae, sometimes with perforations; in addition, for G. resinaceum ellipsoid chlamydospores with smooth surface were observed. Mycelium of G. tsugae was found to have two kinds of clamp connections, single clamps with or without anastomoses while in mycelium of G. oregonense single clamp connections and occasionally swollen hyphae were seen. Double clamp connections and those with anastomoses were noticed for the first time in vegetative mycelium of G. carnosum, as well as staghorn hyphae-in the mycelia of G. sinense and G. carnosum.
