Abstract: The hydrolysis of ATP drives virtually all of the energy-requiring processes in living cells. A prerequisite of living cells is that the concentration of ATP needs to be maintained at sufficiently high levels to sustain essential cellular functions. In eukaryotic cells, the AMPK (AMP-activated protein kinase) cascade is one of the systems that have evolved to ensure that energy homoeostasis is maintained. AMPK is activated in response to a fall in ATP, and recent studies have suggested that ADP plays a… Show more

“…Hence, pharmacologic activation of AMPK would be advantageous in treating different endocrine-related tumors, including prostate and breast cancers (35)(36)(37). However, challenging this view, other studies have emerged that lead to the opposite conclusion, stating that inhibition of AMPK reduces cancer growth (35,36). In particular, the malignant human prostate tissue was shown to display a higher AMPK activity than normal tissue cells.…”

“…AMPK is an energy-sensing serine/threonine kinase consisting of an a-catalytic subunit and regulatory b-and g-subunits. It is activated under conditions of metabolic stress, such as glucose deprivation, hypoxia, and strenuous exercise that deplete intracellular ATP and increase AMP (35). A number of findings suggest that AMPK-mediated reduction in anabolic pathways may ultimately lead to reduced tumor growth.…”

“…AMPK signaling is also linked to the PI3K pathway (Tao et al 2010), which is an important regulator of cell survival (Chen et al 2010). AMPK signaling has, therefore, been investigated as a potential therapeutic target for the treatment of diseases with aberrantly activated PI3K signaling, such as carcinomas (van Veelen et al 2011, Carling et al 2012, Martelli et al 2012. To date, however, to our knowledge no studies have investigated whether AMPK may play a role in adenomyosis.…”

“…AMPK activation by the upstream AMPK kinase LKB1 causes regulation of multiple branches of cellular metabolism that may be important for limiting cancer cell proliferation (Mihaylova and Shaw, 2011). This involves the inhibition of energy‐consuming processes such as fatty acid and protein biosynthesis, while increasing catabolism by promoting autophagy and fatty acid oxidation (Carling et al ., 2012; Faubert et al ., 2015; Mihaylova and Shaw, 2011). In addition to direct regulation of key metabolic checkpoints, AMPK may also inhibit tumorigenesis through upregulation of transcription factors such as the tumor suppressor p53, which is mutated in approximately 80% of all cancers (He et al ., 2014; Jones et al ., 2005; Lee et al ., 2012).…”
