Amoxapine-Loaded Solid Lipid Nanoparticles with Superior Preclinical Pharmacokinetics for Better Brain Delivery: LC-MS/MS and GC-MS Analysis

Abstract: The tricyclic antidepressant amoxapine (AMX) has been reported for a rapid onset of action compared to other cyclic antidepressants. It has very low solubility and bioavailability due to first-pass metabolism. Therefore, we planned to develop solid lipid nanoparticles (SLNs) of AMX using a single emulsification method to increase its solubility and bioavailability. HPLC and LC-MS/MS methods were developed further to quantify AMX in the formulation, plasma, and brain tissue samples. The formulation was studied … Show more

“…The data were collected using control panel software connected to a 21CFR part 11 enabled central server. [38][39][40] The mobile phase was composed of water and acetonitrile in isocratic elution (50:50) with flow rates of 0.7 mL min −1 for OS and 1.2 mL min −1 for NS and ES. The wavelength for the analysis was kept at 212 nm for the three compounds.…”
