Altered molecular pathways in melanocytic lesions

Abstract: To identify gene expression changes in melanocytic lesions, biopsies from 18 common nevi (CMN), 11 dysplastic nevi (DN), 8 radial and 15 vertical growth phase melanomas (RGPM, VGPM), and 5 melanoma metastases (MTS) were analyzed using whole genome microarrays. The comparison between CMN and RGPM showed an enrichment of Gene Ontology terms related to inter and intracellular junctions, whereas the transition from RGPM to VGPM underlined the alteration of apoptosis. Upregulation of genes involved in dsDNA break r… Show more

“…Reverse Transcriptase quantitative Polymerase Chain Reaction (RT-qPCR) was performed as previously described [19] to validate a subset of differently expressed transcripts (CXCL3, GSS, FBXL18 and DUSP1) identified by microarray analysis on fresh samples. Primers sequences are available in Supplementary Table 1.…”

“…Four independent experimental settings were analyzed using the two-color comparative hybridization procedure in three cases and the one color protocol in one. mRNA amplification and labeling was followed by hybridization on 8×60K Human Whole Genome Oligo Microarrays (Agilent Technologies, Santa Clara, CA, USA) with technical replication (details in [68]). Images were analyzed using Feature Extraction software v10.5 (Agilent Technologies) and raw data processed within R statistical environment, using the limma library (http://www.bioconductor.org/packages/2.11/bioc/html/limma.html).…”
