Allelotypic and cytogenetic characterization of chemically induced mouse mammary tumors: High frequency of chromosome 4 loss of heterozygosity at advanced stages of progression

Abstract: Loss of heterozygosity (LOH) is one of the most common genetic abnormalities in cancer. To define the role of LOH and chromosomal abnormalities at various stages of mouse mammary cancer progression, we analyzed the allelotypes and karyotypes of primary mammary tumors induced in CD2F, mice by two basic protocols, the classical multiple-dose 7,12-dimethylbenz[a]anthracene (DMBA) protocol and a novel protocol of combined medroxyprogesterone acetate (MPA) and DMBA. The advantage of the latter protocol is that its … Show more

“…Rat chromosome 5 shows high homology to mouse chromosome 4 [21,23]. As previously stated, genetic alterations of mouse chromosome 4 are found in a variety of mouse tumors [1][2][3][4][5]. The rat genetic marker D5Mit4, which showed AI in our study, is located within 10 cM of the tyrosine phosphatase 1 gene.…”

Exaggerated anticipatory anxiety is common in social anxiety disorder (SAD). Neuroimaging studies have revealed altered neural activity in response to social stimuli in SAD, but fewer studies have examined neural activity during anticipation of feared social stimuli in SAD. The current study examined the time course and magnitude of activity in threat processing brain regions during speech anticipation in socially anxious individuals and healthy controls (HC). Method Participants (SAD n = 58; HC n = 16) underwent functional magnetic resonance imaging (fMRI) during which they completed a 90s control anticipation task and 90s speech anticipation task.

“…Published data reported that these primary tumors are thought to exhibit a low frequency of LOH (loss of heterozygocity) and no apparent genetic instability. 31,32 Because of the inconsistency of our results to published data it was necessary to further investigate the ploidy and chromosome composition of these tumor samples.…”

“…Centrosome anomalies and Aurora A overexpression can coincide with errors occurring during cell cycle and cell division, and are therefore a good indicator for genomic instability and aneuploidy. Published data reported that these primary tumors are thought to exhibit a low frequency of LOH (loss of heterozygocity) and no apparent genetic instability 31, 32. Because of the inconsistency of our results to published data it was necessary to further investigate the ploidy and chromosome composition of these tumor samples.…”

“…Most of the tumors were lobular in morphology and progressively ceased to express receptors after a few in vivo passages. In mice, progestins can act as cocarcinogens together with chemical initiators, such as N-methyl-N-nitrosourea (35) and dimethylbenzanthracene (36). Recent studies in PR knockout mice confirmed the role of PR on dimethylbenzanthracene-induced tumorigenesis (37).…”
