Allelic variation and genetic linkage of avian microsatellites in a new turkey population for genetic mapping

Abstract: Efforts to build a comprehensive genetic linkage map for the turkey (Meleagris gallopavo) have focused on development of genetic markers and experimental resource families. In this study, PCR amplification was attempted for 772 microsatellite markers that had been previously developed for three avian species (chicken, quail and turkey). Allelic polymorphism at 410 markers (53.1% of total examined) was determined by genotyping ten individuals (six F1 parents and four grandparents) in a new resource p… Show more

“…species ranged from 16.23 to 62.89% (Moore et al 1991;Kemp et al 1995;Fields and Scribner 1997;Baratti et al 2001;Kayang et al 2002;Reed et al 2003;Kim et al 2004). Studies show that it was difficult to screen microsatellite markers for duck by cross-species amplification from chicken loci (Huang et al 2005).…”

“…The structure of the genomes of the chicken and turkey are likely to be very similar, with at least one major chromosomal difference (Schmid et al, 2000). Linkage relationships and marker order as determined for several chicken markers genotyped in the turkey, including ADL0107 and ADL0315, appear to be comparable to those observed in the chicken Reed et al, 2003). Based on these data, MYL1 in the turkey is assigned to a linkage group corresponding to a region of GGA7.…”

“…Two-point linkage analysis was performed with all previously genotyped markers . Significant pairwise linkage relationships were found between MYL1 (F147) and two other markers, ADL0107 and MNT-ADL0315 (MNT-ADL0315 corresponds to chicken locus ADL0315 but utilizes primers designed from turkey DNA sequence, Reed et al, 2003). Both of these loci have been mapped in the chicken to chromosome 7 (GGA7, Schmid et al, 2000).…”

“…Several procedures for microsatellite library enrichment have been developed to improve the efficiency of microsatellite isolation. They can be classified according to the capture technique: 1) streptavidin-coated magnetic beads; 2) microsatellite probes attached to small nylon membranes; and 3) other infrequently used procedures, such as the use of the dut ung strain of E. coli or the magnetic capture of phage DNA (Reed et al, 2003).…”

“…For linkage and comparative genetic mapping, parentage testing, population genetics, and phylogenetic studies in Cervidae, we isolated and characterized polymorphic microsatellite markers in sika deer. After reading the literature pertaining to microsatellite libraries, we decided to utilize the strong affinity between biotin and streptavidin to construct the microsatellite library (Reed et al, 2003). Using this approach, we isolated and characterized a number of microsatellite markers.…”
