Alkaline Phosphatase-Triggered Simultaneous Hydrogelation and Chemiluminescence

Abstract: Chemiluminescence (CL) has a higher signal-to-noise ratio than fluorescence, but the use of CL to track an enzyme-instructed self-assembly (EISA) process has not been reported. In this work, by coincubation of the hydrogelator precursor Fmoc-Phe-Phe-Tyr(HPO)-OH (1P) and the CL agent AMPPD (2) with alkaline phosphatase (ALP), we employed CL to directly characterize and image the simultaneous EISA process of 1P. Hydrogelation processes of 1P with and without 2 and the CL properties of 2 with and without 1P under… Show more

“…Fluorescein (Fl) was employed as the competitive dye on accounts of its strong binding with GC5A‐6C, the high brightness itself and the drastic complexation‐induced quenching of fluorescence . Alkaline phosphatase (ALP) was engaged in degrading PLP to PL and P . Therefore, this combinational system executes the ability to detect PLP.…”

“…There are three kinds of self‐luminescence imaging approaches: bioluminescence, chemiluminescence, and afterglow imaging (Figure b). Bioluminescence relies on the enzymatic reaction to generate photons, and thus involves the enzyme and substrate, while chemiluminescence is produced by chemical reactions between substrates and often reactive species . Different from bioluminescence and chemiluminescence triggered by addition of a substrate into the system, afterglow imaging requires preirradiation of light to generate the chemical or energy defects and detects photons released from those defects after cessation of light irradiation (Table ) …”

“…By GSH reduction, the hydrophilic EE sequence was removed, leading to the formation of curcumin-based supramolecular nanofibers for use as radiosensitizers. Liang and coworkers 107, 108 design a precursor Cys(SEt)-Glu-Tyr(H 2 PO 3 )-Phe-Phe-Gly-CBT that is responsive to extracellular ALP and intracellular GSH in tumor site for yielding cyclic amphiphilic 2D building blocks. Interestingly, the precursor molecules cannot enter the cells, but the converted cyclic amphiphilic 2D molecules can be efficiently internalized by cells and locally self-assemble into the nanofibers with enhanced mechanical strength, highlighting great potential of smart nanotheranostics design.…”
