Aliphatic Substitution of o-Carboranyl Phenols Enhances Estrogen Receptor Beta Selectivity

Abstract: The two subtypes of estrogen receptor (ER), ERα and ERβ, differ greatly in expression pattern and biological functions, and ERβ-selective ligands candidates to treat immune-related disorders. ERβ-selective ligands have mostly been designed based the idea of introducing a substituent that interferes sterically with the ligand's interaction with Met421 to selectively decrease the affinity for ERα (the equivalent residue in ERβ is Ile373). Therefore, we designed and synthesized a series of carboranyl phenol deriv… Show more

“…In particular, n -propyl-substituted o -carborane 42 (Fig. 9) shows an interesting 7.4-fold beta selectivity in ER-binding assays, with RBA values of 11.8% on ERα and of 87% on ERβ [118]. Most importantly, this compound displayed only moderate estrogenic activity in MCF-7 cells and, therefore, may constitute a promising starting point for the development of ERβ-selective carborane-containing ligands.…”

“…29 More recently, the same group worked on the synthesis and biological evaluation of carboranyl-based compounds with selective affinity for specific ER subtypes. 30 Nuclear ERs exist in fact as two major subtypes, a and b, which are composed of several domains: DNA binding, hormone binding and activation of transcription domains. A variety of nuclear responses are mediated through the binding of estrogens (17b-estradiol, estrone, estriol, estetrol), or estrogen analogues, to the ligand binding domain (LBD), which transactivates gene expression upon ligand binding.…”

“…Owing to these distinctive properties, carboranes have been regarded as one of the most excellent organic groups to improve performance of organic polymers with remarkable thermal stability . Therefore, carboranes were incorporated into many traditional polymers to improve their thermal stability including epoxy resins, phenolic resins, polyurethanes, organic silicone polymers, benzoxazine resins, polyimides, and so forth.…”
