Agrobacterium Tumefaciens Mediated Transformation of rolC::Hd3a-GFP in Black Rice (Oryza Sativa L. cv. Cempo Ireng) to Promote Early Flowering

Abstract: The purpose of this study was to transform rolC::Hd3a-GFP by Agrobacterium tumefaciens on black rice (Oryza sativa L. cv. Cempo Ireng). The callus formation were induced from scutellum using two different media, 2N 6 and 2,4-dichlorophenoxyacetic acid (2,4-D). PCR analysis using specific primers for Hd3a and hpt was performed to determine the stability of rolC::Hd3a-GFP construct before co-cultivation of callus and Agrobacterium tumefaciens. This study indicated that black rice callus induction on 2N 6 medium … Show more

“…In order to promote early flowering in Cempo Ireng, insertion flowering gene Hd3a can be done by transformation rolC::Hd3a-GFP mediated by Agrobacterium tumefaciens into callus of the Cempo Ireng [34,35]. According to Tamaki et al [36], the expression of Hd3a in japonica rice sub-species was controlled by rolC promoter which accelerate the flowering period.…”

“…Thus, we should implement efforts to shorten this flowering period to enhance yield. To overcome this problem, a preliminary study has been initiated in order to investigate and understand flowering behaviour [53]. Recent studies suggest that 'Cempo Ireng' black rice grown under neutral day conditions has a flowering pathway conserved with a rice model grown under SD/LD condition.…”
