Age-Dependent Vulnerability to Oxidative Stress of Postnatal Rat Pyramidal Motor Cortex Neurons

Abstract: Oxidative stress is one of the main proposed mechanisms involved in neuronal degeneration. To evaluate the consequences of oxidative stress on motor cortex pyramidal neurons during postnatal development, rats were classified into three groups: Newborn (P2–P7); infantile (P11–P15); and young adult (P20–P40). Oxidative stress was induced by 10 µM of cumene hydroperoxide (CH) application. In newborn rats, using the whole cell patch-clamp technique in brain slices, no significant modifications in membrane excitabi… Show more

“…According to previous studies, oxidative stress vulnerability increases with age, with adults being more vulnerable and juveniles being partially resistant (30,31). In the present study, effects of 1 mM H 2 O 2 on GnRH neurons were studied in three groups according to age: juvenile, 8 to 25 postnatal days (PND); peripubertal, 26 to 45 PND; and adults, more than 60 PND.…”

“…However, unlike adults, most immature GnRH neurons were stimulated by the same concentration of H 2 O 2 . According to previous studies, oxidative stress vulnerability increase with age, with young rats being more resistant to ROS than adults (30). Furthermore, H 2 O 2 has both excitatory and inhibitory effects on neuronal excitability depending on neuronal population and brain location (43).…”

“…Using a micromanipulator (MP-225, Sutter Instrument, CA, United States), micropipettes were visually placed in the motor cortex area. Within this region the pyramidal neurons were identified based on their characteristic morphology ( Pardillo-Díaz et al, 2016 ; Carrascal et al, 2020 ). An amplified (MultiClamp 700B, Axon Instruments, Molecular Devices, Sunnyvale, CA, United States) was also used to achieve the whole-cell patch clamp configuration.…”

“…Therefore, based on all our previous results, it is reasonable to hypothesize that damage caused by oxidants such as CH mainly contributes to neuronal dysfunction in aged brains, and therefore, neurodegenerative diseases such as ALS are more likely to occur in the older population [ 140 ].…”
