Advances in isolation and detection of circulating tumor cells based on microfluidics

Abstract: Circulating tumor cells (CTCs) are the cancer cells that circulate in the peripheral blood after escaping from the original or metastatic tumors. CTCs could be used as non-invasive source of clinical information in early diagnosis of cancer and evaluation of cancer development. In recent years, CTC research has become a hotspot field wherein many novel CTC detection technologies based on microfluidics have been developed. Great advances have been made that exhibit obvious technical advantages, but cannot yet s… Show more

“…Recently, Strep-tag technique is studied to isolate CTCs with 79% of capture efficiency using EpCAM, HER2, EGFR biomarker and magnetic beads, which showed CTC enrichment under external magnetic field with very fast isolation 32 . This process relies on an expensive EpCAM biomarker, which leads expensive detection and limits its wide-spread use [33][34][35] . Wen et al proposed similar magnetic nanospherical modules, prepared by an emulsifier-free polymerization method coupled with layer-by-layer assembly to capture rare tumor cells 36 .…”

“…However, the information that could come from the investigation of these cells is enormous. Therefore, it is worth the effort to find new methods to isolate and study them [ 78 ]. Characterizing these cells could give information about the stage of a tumor.…”

“…Microfluidic methods allow for gentle capturing of live CTCs so that further downstream analysis can be performed using cellular, microscopic, or molecular approaches, to meet the clinical or research need. Microfluidic based CTC-capture devices can be modified to exploit physical and/or biological properties between CTCs and the background cells, thus enabling isolation 24. On the basis of microfluidic technique, CytoSorter® CTC-capture system employs an antibody-immobilized chip for immune capture of CTCs, which are then verified by immunofluorescence staining for PanCK, CD45, and DAPI.…”
