Abstract: Adiponectin has been shown to stimulate fatty acid oxidation and enhance insulin sensitivity through the activation of AMP-activated protein kinase (AMPK) in the peripheral tissues. The effects of adiponectin in the central nervous system, however, are still poorly understood. Here, we show that adiponectin enhances AMPK activity in the arcuate hypothalamus (ARH) via its receptor AdipoR1 to stimulate food intake; this stimulation of food intake by adiponectin was attenuated by dominant-negative AMPK expression… Show more

“…24 Recently, we showed that adiponectin also centrally regulates energy expenditure and food intake. 28 We also identified AdipoRs (AdipoR1 and R2) by expression cloning, 15 and found that AdipoRs are also decreased in obesity. 26 Overexpression by adenoviruses suggested that strategies to increase AdipoRs should serve as treatment strategies for insulin resistance, metabolic syndrome, diabetes and atherosclerosis.…”

“…28 After feeding, serum leptin levels increase, which may lead to decreased hypothalamic AMPK activity and suppress food intake. In contrast, during fasting, serum adiponectin levels and AdipoR1 expressions in the arcuate hypothalamus increase, which may lead to increased hypothalamic AMPK activity and promote food intake.…”

“…AdipoR1 and AdipoR2 are both abundantly expressed in the hypothalamus, particularly in the arcuate nucleus and paraventricular hypothalamus 22. Intracerebroventricular administration of APN in a mouse model of T2DM was shown to decrease body weight, mostly through stimulating energy expenditure 23.…”

“…Whereas APN receptors are predominantly expressed in the hypothalamus,22 there seem to be extensive changes in APN levels in other brain regions in neurodegenerative diseases. Currently, the mechanism through which the expression pattern of APN is distinct from those of APN receptors remains unclear.…”
