Adenovirus-mediated expression of BmK CT suppresses growth and invasion of rat C6 glioma cells

Abstract: BmK CT, one of the key toxins in the venom of the scorpion, Buthus martensii Karsch, can interact specifically with glioma cells as a chloride channel blocker and inhibit the invasion and migration of those cells via MMP-2. A recombinant adenovirus, Ad-BmK CT, was constructed and characterized by in vitro and in vivo studies, using MTT cytotoxicity assay and the glioma C6/RFP (red fluorescence protein)/BALB/c allogeneic athymic nude mice model, respectively. The adenovirus-mediated expression of BmK CT display… Show more

“…BmKCT interacts specifically with human glioma (SHG-44) cells, but not with normal astrocytes, as a Cl‒ channel blocker [54] and inhibits the invasion and migration of rat glioma (C6) cells by antagonizing MMP-2 [55]. Similarly, the recombinant adenovirus-produced BmKCT, Ad-BmKCT, reduced rat glioma (C6) cell viability in vitro and the growth and metastasis of xenografted rat glioma (C6) tumors in female athymic nude mice following intratumoral injection of Ad- BmKCT (100 µL, 1010 viral particles, every five days) [56].…”

“…Targeting BmK CT to C6 glioma cells specifically through this delivery system had been developed by them. Thus, the replication-defective recombinant adenovirus Ad-BmK CT could provide a powerful delivery system to treat glioblastoma 52 Moreover, improved antitumor effects on the subcutaneous U251 or SHG-44 tumors of the nude mouse models had been achieved by the CTX-Onc conjugate compared to the CTX+Onc control. These results were indicative of the promotion of the tumor targeting of Onc through the chemically reversible conjugated CTX.…”
