“…Serious cardiovascular events have been reported in temporal association with sildenafil use. 1 As cardiovascular and cerebrovascular diseases share many similarities, the possible association of stroke and sildenafil has been speculated. Supporting this association are recently published cases of anterior ischemic optic neuropathy (AION) associated with sildenafil use.…”

“…A number of serious cardiovascular adverse events have been associated with its use, including myocardial infarction and sudden cardiac death in adults with coronary artery disease [2]. These adverse events were attributed to the systemic vasodilating effect of Sildenafil resulting in systemic hypotension in patients receiving nitrates.…”

“…The most common adverse effects when using tadalafil, along with other PDE5 inhibitors are headache, indigestion, back pain, muscle aches, flushing, and stuffy or runny nose. Although there is no evidence of hypercoagulation states being associated with the use of PDE5 inhibitors, several studies [3][4][5][6][7][8] suggest that PDE5 inhibitors may, in rare cases, cause arterial and venous thrombotic or embolic events as summarized in Table 1. However, Zusman et al [13] performed a retrospective investigation on the incidence of serious cardiovascular adverse events, including stroke and myocardial infarction, and the outcome was the same for patients treated with sildenafil or placebo.…”

“…Well-known adverse reactions to PDE5 inhibitors, such as headache, flushing, and nasal congestion, demonstrate that vasodilation is not limited to the corpus cavernosum [2]. However, several studies [3][4][5][6][7][8] have also suggested that PDE5 inhibitors may, in rare cases, cause arterial and venous thrombotic or embolic events, even though to date there is no direct association between PDE5 inhibitors and hypercoagulation states. We describe the first case of pulmonary embolism (PE) in a patient with protein C deficiency that had just begun using tadalafil for ED.…”
