Activity of mannose‐binding lectin in centenarians

Abstract: We analyzed MBL2 gene variants in two cohorts of centenarians, octo-and nonagenarians and in the general population, one from Sardinia island (Italy), recruited in the frame of the AKea study, and another from Campania (southern Italy), to search for haplotypes related to longevity. We also assessed in vitro the effect of mannose-binding lectin (MBL) on various human cells at different stage of senescence. The frequency of high and null activity haplotypes was significantly lower and the frequency of intermedi… Show more

“…A correct balance between pro-inflammatory and anti-inflammatory networks, which avoids either a too strong or a too weak reaction to inflammatory stimuli, is crucial for successful aging. This notion fits with recent data showing that among different polymorphisms of the mannose binding lectin (MBL, a well known PRR), only those with intermediate levels of activity were associated with extreme longevity (Tomaiuolo et al, 2012).…”

“…HNECs can be effectively cultured up to 15 days, as we also demonstrated in a previous study in which we assessed the effect of mannose binding lectin on several types of cells 11 . So far, only a few studies have been performed on the use of human ex vivo models for CF.…”

“…The sex and age distribution of this group (see Table 1) matched the anagraphic distribution of subjects in southern Italy. For most of the subjects from the general population, DNA samples were already available in the bank of biological samples from the institutions involved in the study, and are currently used collectively for the study of gene variants associated with inherited diseases [15,16]. …”
