Activity of Different Desoximetasone Preparations Compared to Other Topical Corticosteroids in the Vasoconstriction Assay

Abstract: Introduction: We report on a double-blind, vehicle-controlled, single-center confirmatory study with random assignment. The purpose of the study was to investigate the topical bioavailability of different topical corticosteroid formulations in healthy human beings focussing on desoximetasone (DM). Materials and Methods: Two DM 0.25% formulations [ointment (DM-o) and fatty ointment (DM-fo, water-free); class III corticosteroids], the corresponding active ingredient-free vehicles and three comparators of differe… Show more

“…Desoximetasone is a synthetic topical corticosteroid used to treat mild to moderate psoriasis effectively. Desoximetasone has been used in various semisolid dosage forms such as creams, ointments, and gels; however, all of these formulations are limited in terms of controlled release applications, since no product with sustained release is currently commercially available [ 27 , 28 , 29 ].…”

“…It is based on the property of corticosteroids to induce vasoconstriction represented by blanching of the skin [19][20][21] . The vasoconstrictive strength is known to correlate strongly with the clinical efficacy of topical glucocorticoids as determined in clinical treatment trials [22] .…”

“…A first approach to the development of an in vitro test system was based on the inhibition of interleukin (IL)-1 synthesis in normal human keratinocytes and fibroblasts (13). The results were in close correlation with those from studies in human volunteers, when comparing GCs which varied in safety but were similar in skin blanching (16). The latter effect is used for the estimation of clinical potency (17).…”
