Acacetin Blocks Kv1.3 Channels and Inhibits Human T Cell Activation

Abstract: Backgrounds/Aims: Acacetin, a natural flavonoid compound, has been proven to exert anti-inflammatory and immunomodulatory effects. Kv1.3 channels, highly expressed in human T cells, are attractive therapeutic targets to treat inflammatory and immunological disorders. The present study was designed to characterize the inhibition of Kv1.3 channels by Acacetin in human T cells and examine its role in T cell activation. Methods: Whole-cell patch-clamp was applied to record the Kv1.3 and KCa currents in … Show more

“…39, 40 In brief, NRVM and H9C2 cells were loaded with 2  μ M Fluo-4/AM (Molecular Probes, Carlsbad, CA, USA, #F14201) for 30 min, washed three times with HEPES buffer that contained (in mM) 130 NaCl, 4.7 KCl, 2.0 CaCl 2 ,1.2 MgSO 4 , 1.2 KH 2 PO 4 , 10 HEPES and 11 glucose at pH 7.4. Cells in 96-wells plate were illuminated at 488 nm and fluorescence emissions at 525 nm were captured with the Enspire Multimode Plate Reader (PerkinElmer, Boston, MA, USA).…”
