“…However, we did not see multiplicated ventral fin folds in any of the morphants, although this is a sensitive indicator of increased BMP signaling and is a consistent feature of the ventralized din and ogon mutants . We did observe narrowing of the anterior nervous system, but morpholinos can induce nonspecific toxic effects in zebrafish (Heasman, 2002), including widespread cell death and neural degeneration (Braat et al, 2001;Lele et al, 2001). In support of this possibility, increased apoptosis occurs in the brains of tsg1 morphants (Little and Mullins, 2004) and is not seen in ventralized din mutants .…”

“…In medaka, dnd RNA utilizes particle partition as a mechanism for asymmetric segregation of PGC fate decision during early embryogenesis (Hong et al, 2016). However, knockdown of vasa did not affect PGC survival in zebrafish and medaka (Braat et al, 2001;. Based on our present study and previous reports in gibel carp, we have identified three germ cell markers vasa, dazl and dnd.…”

“…Testes require at least two weeks to develop, and oocytes can mature over several generations, therefore our treatment might not be long enough to assess the potential role of vasa in germline development. Conversely, vasa knockdown in different species shows disparate phenotypes, ranging from no defects [zebrafish (Braat et al, 2001) and flatworms (Pfister et al, 2008)] to sex-specific defects on male gamete formation, but not female [mice (Tanaka et al, 2000)], to defects in migration but not in the identity of the germ cell precursors [medaka (Li et al, 2009)]. Thus, it would not be surprising if vasa was shown not to be required for germline formation; however, its expression can still be used to identify precursors, as has been shown in other animals.…”
