QTL Analysis for Resistance to Phytophthora Blight (Phytophthora capsici Leon.) Using an Intraspecific Doubled-Haploid Population of Capsicum annuum

A doubled-haploid (DH) population (n = 176) obtained by anther culture of an F 1 hybrid between a line susceptible to Phytophthora capsici 'K9-11' (Capsicum annuum L.) and a line resistant to P. capsici 'AC2258' (C. annuum L.) was inoculated with P. capsici. QTL analysis of the resistance was performed using a linkage map consisting of 16 linkage groups (LGs), covering a total distance of 1100.5 cM. Three QTLs were detected on LG1, LG6 and LG7. The QTL with the highest LOD score, detected on LG7, explained 82.7% of the phenotypic variance with a LOD score of 67.02. This QTL was designated as Phyt-1. The nearest marker was an AFLP marker, M10E3-6. The second QTL, designated as Phyt-2, was found on LG1. It explained 6.4% of the phenotypic variance with a LOD score of 2.54. The nearest RAPD marker was RP13-1. The other QTL, designated as Phyt-3, which was found on LG6, explained 5.6% of the phenotypic variance with a LOD score of 2.20. The nearest AFLP marker was M9E3-11. It was confirmed that the lines with a high resistance could be efficiently selected by using two markers, M10E3-6 and RP13-1, simultaneously. The presence of both Phyt-1 and Phyt-2 under homozygous conditions may enable to breed resistant cultivars of sweet pepper. The molecular markers identified in the present study could be useful for marker-assisted selection (MAS) in order to breed sweet pepper cultivars with a high resistance to P. capsici using 'AC2258' as a source of resistance genes.

Greenhouse studies were performed to determine the reactions of 10 "California Wonder" (Capsicum annuum) accessions to the three forms of Phytophthora blight (root rot, stem blight and foliar blight) caused by Phytophthora capsici. Differences in root rot, stem blight and foliar blight severities among accessions were significant. The accessions consistently differentiated into two groups across the three disease syndromes. Simple sequence repeat (SSR) markers showed variability both within and between accessions of California Wonder. The variability in the responses to the three forms of Phytophthora blight does not warrant its usefulness as a standard susceptible control in studies involving the Capsicum-P. capsici pathosystem.

Resistance of one hundred commercialized cultivars of chili pepper to four isolates of Phytophthora capsici was evaluated under controlled environmental conditions. The cultivars are commercialized as resistant (59%) and susceptible (41%) to Phytophthora blight in Korea. Mean disease severities of the cultivars on P. capsici MY-1, KPC-1, JHAI1-7, and KPC-7 isolates were 37, 55, 60, and 74%, respectively. In addition, 38 for MY-1, 48 for KPC-1, 56 for JHAI1-7, and 76 cultivars for KPC-7 showed susceptibility. To P. capsici MY-1, the weakest pathogenicity isolate among them, 59 cultivars represented high resistance. By contrast, only six cultivars showed high resistance to P. capsici KPC-7, the strongest isolate. Furthermore, resistance of most cultivars except for three cultivars was negatively correlated with the virulence of P. capsici isolates. And isolate-specific resistance of the chili pepper cultivars could not be found. Among them, six cultivars showing resistance to all the tested isolates were selected for further study. The development of Phytophthora blight on the six cultivars according to inoculum density (5 × 10 4 to 1.5 × 10 6 sporangia/pot) and incubation temperature (25 to 30°C) after inoculation of P. capsici was tested. Resistance of the cultivars to P. capsici KPC-1 and JHAI1-7, moderately pathogenic isolates, was hardly affected. But to KPC-7 isolate, the highly resistant cultivars showed susceptiblility or moderate resistance when the seedlings were inoculated with inoculum density of 1.5 × 10 6 sporangia/pot and incubated at 28 to 30°C. From these results, it is likely that resistance of chili pepper cultivars to Phytophthora blight is affected by the virulence of P. capsici isolate.
