A Probiotic Spore‐Based Oral Autonomous Nanoparticles Generator for Cancer Therapy

Abstract: Spores, the dormant life forms of probiotics, can germinate to metabolically active vegetative cells with the disintegration of their hydrophobic protein coat in the intestinal microenvironment, which provides the possibility for the formation of nanoparticles (NPs) in vivo. Inspired by the natural physiological process of spores, herein, an oral autonomous NPs generator is developed to overcome the spatially variable gastrointestinal tract environment and multibiological barriers. Spores modified with deoxych… Show more

“…The application of probiotic spores as a supply chain for chemotherapeutic drugs was recently proposed. In the autonomous production of nanoparticles in the gastrointestinal tract, Song et al developed modified deoxycholic acid spores packed with doxorubicin and sorafenib [224]. Such a strategy targets the safety of medications in order to boost bioavailability in oral administration.…”

“…Peripheral immune cells are a class of immune cells including lymphocytes, antigen presenting cells (APCs), mast cells, and other immune cells, which reside in the peripheral immune organs, and are mainly responsible for recognizing antigens and participating in the immune response. Within the peripheral immune organs, the induction of anti-tumor immunity is a multi-step process, which includes the recognition and presentation of tumor antigen by APCs, activation of antigen-specific T cells, and attacking of the tumor cells by effector T cells (Song Q. et al, 2019). However, tumor cells can usually escape immunosurveillance through a number of special mechanisms, which largely reduce the body's immune response and ensures the survival of cancer cells (Mohme et al, 2017).…”

“…The use of spores from probiotics have been recently proposed as a delivery system for chemotherapeutic drugs. Song et al [175] produced deoxycholic acid-modified spores to be loaded with doxorubicin and sorafenib as an approach for autonomous production of nanoparticles in the gastrointestinal tract. Such approach envisions drug protection upon oral administration to improve bioavailability.…”
