A Polymorphism in the Serotonin Transporter Gene Regulatory Region and Frequency of Migraine Attacks

Abstract: This polymorphism does not appear to be involved in a genetic predisposition to the disease but may affect the frequency of attacks in patients with migraine. These findings may contribute to our understanding of factors that influence the clinical severity of migraine.

“…While there is currently no clear evidence for 5-HTT polymorphisms being associated with migraine without aura [17,21,28,40], previous genetic studies pointed to a possible link between carriers of the short allele of the 5HTTLPR (5-HT gene linked polymorphic region) and migraine with aura [7,25]. So far, investigations which focused on a possible correlation between the 5-HTTLPR polymorphism and the availability of SERT-radioligands failed to show a simple genotypephenotype relationship [36].…”

“…Twelve studies, which included a total of 1502 cases and 1958 controls, dealt with the association between 5-HTTLPR and migraine [12][13][14][31][32][33][34][35][36][37][38][39]. Four studies, involving 495 cases and 729 controls, examined the risk of 5-HTT VNTR in migraine [11,36,37,40].…”

“…Of studies investigating the 5-HTTLPR polymorphism, eight were performed on Caucasian populations [12][13][14]32,33,35,37,38] and four on Asian populations [31,34,36,39]. Nine studies examined patients with MA and MO [12,13,31,[33][34][35][37][38][39], one with MA alone [14] and two with MO alone [32,36]. In studies of 5-HTT VNTR polymorphism, three were carried out on Caucasians [11,37,40] and one was on Asians [36].…”

“…Polymorphisms in the serotonin transporter gene (5HTTLPR) were identifi ed, with two polymorphic regions noted-one with a short-based pair region of 484 based pairs, and one with a long-based pair region of 428 polymorphisms [ 32 ]. Patients with migraine were compared with controls and could be classifi ed as having two short regions, two long regions, or a mixture.…”
