A novel IL‐23p19/Ebi3 (IL‐39) cytokine mediates inflammation in Lupus‐like mice

Abstract: Interleukin-12 family cytokines have emerged as critical regulators of immunity with some members (IL-12, IL-23) associated with disease pathogenesis while others (IL-27sue injury by promoting the expansion of regulatory B and T-cell subsets [2,3].Discovery of IL-23 in 2000 [4] led to the reevaluation of IL-12 and IL-23 in autoimmune diseases. For example, therapeutic targeting of IL-12p40 decreases pathology in many mouse models of autoimmune diseases [5], while disease is exacerbated in IL-12p35-deficient m… Show more

“…For the sake of completeness, it should be mentioned that very recently another family member, IL-39, has been described. IL-39 is composed of IL-23p19 and EBI3 heterodimer, is secreted by activated B lymphocytes and seems to play a pathogenic role in mouse models of systemic lupus erythematosus [70,71], but no data on RA or other diseases are available so far. These cytokines transduce the signal through unique pairings of 5 receptor chains: IL-12Rβ1, IL-12Rβ2, IL-23R, IL-27Rα (or WSX-1) and gp130.…”

“…This suggests that IL-23p19 and EBI3 may function as a novel IL-12 family cytokine. Indeed, these two subunits were very recently reported to form IL-39, which was shown to promote neutrophil expansion in lupus-prone mice [64,65]. Therefore, IL-39 may play a role downstream of TLR3 in wound healing responses in the skin by promoting neutrophil-mediated responses at the site of injury.…”

“…It signals through four receptor complexes: IL-12Rβ2/gp130, IL-12Rβ2/IL12Rβ2, gp130/gp130, and IL-12Rβ2/ WSX-1 15,16 . Recently discovered IL-39 consists of p19 and EBI3 chains and signals through IL-23R/gp130 17 . The heterodimer p28/p40, tentatively called IL-Y, has been demonstrated to antagonize the signaling by IL-12 and IL-27 18 .…”

“…IL-35 is mainly produced by FoxP3+ regulatory T (Treg) cells, but can also be produced by gdT cells, CD8+ T-cells, and placental trophoblast 24 . In addition, it has been demonstrated that activated B cells secrete IL-39 17 .…”
