Abstract. We examined the effects of Ca 2+-channel blockers on sugar cataract formation in streptozotocin (65 mg / kg, i.v.)-induced diabetic rats that were given 5% D-glucose as drinking water. The diabetic rats were treated with an L-type Ca 2+-channel blocker, nifedipine or verapamil, for 9 weeks from the 3rd day of streptozotocin injection. Using the full lens images of the horizontal plane captured with the new digital camera system that we developed recently, the cataract formation was quantitatively assessed in parallel with the conventional scaling method. In the animal model of diabetes mellitus, the cataracts at the peripheral region of the lens were detected 2 weeks after induction of hyperglycemia and progressed depending on the length of the diabetic period. The majority of them developed mature cataracts after 9 weeks of hyperglycemia. Nifedipine slowed the progression rate of diabetic cataracts without affecting the period of time required for the onset of this disease, whereas verapamil had no significant inhibitory effect on the diabetic cataract. These findings suggest that nifedipine may be considered as a candidate drug to suppress the progression of diabetic cataracts.

Background: The aim of our study was to compare metabolic (oxygen saturation; %) and anatomical parameters (retinal vessel diameter; lm) of retinal vessel oximetry (RO) in patients with cataract formation against those of healthy controls with clear lenses. Methods: A total of 96 eyes of 62 subjects were examined: 51 eyes from 33 cataract patients (mean age: 64.8y) were compared to 45 eyes from 29 controls with clear lenses (mean age: 61.5y). RO was performed with the oxygen saturation measurement tool from the RVA (IMEDOS Systems UG). The oxygen saturation in all major peripapillary retinal arterioles (A-SO 2 ) and venules (V-SO 2 ) was measured, and their difference (A-V SO 2 ) was calculated. In addition, the corresponding diameter in retinal arterioles (D-A) and venules (D-V) was evaluated. Cataract formation was graded according to the Lens Opacities Classification System III (LOCS III). Oximetry data were compared with the grade of cataract formation within both groups. For statistical evaluation, ANOVAbased linear mixed-effects models were calculated (SPSS â , pairwise comparisons: Bonferroni-corrected; p < 0.05). Results: Cataract eyes showed significantly lower A-SO 2 and A-V SO 2 values (mean AE SD 92.52 AE 9.80% and 28.56 AE 9.80%), when compared to healthy controls (95.47 AE 4.48% and 34.8 AE 7.08%; p = 0.046 and p = 0.001). Within the cataract group, cortical opacities showed significant interactions with the A-SO 2 , V-SO 2 and the A-V SO 2 parameters (p = 0.027; p = 0.002; and p = 0.026, respectively). Conclusions: These data indicate that the cataract-induced light scatter influences optical retinal oxygen measurements. Cortical opacities showed the highest influence on RO measurement when compared to nuclear opacification, nuclear colour and posterior cataract formation.

Probucol Slows the Progression of Cataracts in Streptozotocin-Induced Hyperglycemic Rats

We examined the effect of probucol, an antihyperlipidemic drug with potent antioxidant properties, on cataract formation in streptozotocin (STZ)-induced hyperglycemic rats that were given 5% D-glucose as drinking water. Probucol treatment was initiated immediately after the induction of hyperglycemia was confirmed. Using full horizontal-plane lens images captured with an original digital camera system, the opacity of central region of lens was assessed by measuring the opaque area in the region. Central opacities were detected after 3 weeks of hyperglycemia, and progressed in a time-dependent manner. The majority of STZ-induced hyperglycemic rats developed severe cataracts after 9 weeks of hyperglycemia. Probucol slowed the progression of cataracts in a dose-dependent manner. Levels of sorbitol and protein carbonyls in lenses of STZ-induced hyperglycemic rats were higher than those of control rats. Probucol suppressed the increase in protein carbonyls, but not of sorbitol, in lenses of STZ-induced hyperglycemic rats. Probucol had no significant effect on increases in plasma concentrations of glucose, total cholesterol, and triglyceride observed in STZ-induced hyperglycemic rats. These results suggest that probucol slows the progression of sugar cataracts, independent of its lipid-lowering effects. The beneficial effect of probucol on cataracts is partially attributable to the attenuation of oxidative damage to lens proteins.
