A new dimeric neolignan from Magnolia grandiflora L. seeds

Abstract: Bioassay-guided fractionation of the MeOH extract of Magnolia grandiflora seeds resulted in the isolation of a new dimeric neolignan, named bishonokiol A (1), as well as two known neolignans magnolol (2) and honokiol (3). The structures of the compounds were determined on the basis of data obtained using NMR and MS. Bishonokiol A (1) showed potent anti-proliferative activities in four human cancer cell lines, with IC50 values ranging from 5.1 to 7.5 µM. Additionally, bishonokiol A (1) induced apoptosis, as wel… Show more

“…Many in vitro and in vivo investigations offer evidence of the effectiveness of MAG against liver cancer where it is found to increase cell cytotoxicity, repress cell proliferation/cell viability and reduce tumor growth significantly [61,127,129,141,142,143]. MAG induced apoptosis in HepG2 cells by increasing the intracellular level of calcium along with increased translocation of cyt- c from mitochondria to the cytosol and activation of caspase-3, -8, and -9 [57].…”

“…Thus, the search for new anti-cancer agents from natural sources has gained importance in recent years. Bishonokiol A (BHNKA) is a dimeric neolignan isolated from Magnolia grandiflora seeds, and exhibits anti-proliferative activity, induces apoptosis, and inhibits invasion and migration of cancer cells (Figure 1A) (Li et al, 2015;Li et al, 2019). However, the pharmacological mechanism of BHNKA induced breast cancer cell death is still unclear.…”

“…BHNKA was isolated from M. grandiflora, and the purity of BHNKA was higher than 96% by HPLC analysis (Li et al, 2015). Dulbecco's modified Eagle's medium (DMEM) and 0.25% trypsin were purchased from Hyclone (UT, USA).…”

“…M. grandiflora has well‐known uses in American indigenous medicine as bitter tonics, antimalarials, and diaphoretics, [4] as well as in traditional Chinese medicine, in which the use of leaves for the treatment of hypertension has been reported [5] . Previous studies on M. grandiflora have led to the isolation and identification of various kinds of compounds, including alkaloids, [6] sesquiterpenoids, [7–9] sesquiterpene lactones, [10,11] flavonoids, [12] lignans [13] and neolignans [14–16] . Additionally, several studies have focused on the composition of volatile fractions and essential oils of different plant parts.…”
