Abstract: Highlights d A long-form tracrRNA (tracr-L) is a natural single-guide RNA for S. pyogenes Cas9 d tracr-L directs Cas9 to repress CRISPR-Cas expression and mitigate autoimmunity d De-repression of tracr-L enhances immunity to bacteriophage infection by 3,000-fold d tracr-L is conserved in many type II-A CRISPR-Cas systems
