Abstract: Objectives: Acinetobacter baumannii can cause severe nosocomial and community-acquired pneumonia. To study the pathogenesis of A. baumannii and to develop new treatments, appropriate mouse models are needed. Most reported mouse models of pulmonary A. baumannii infection are non-lethal or require mouse immunosuppression to enhance infection. These models are not suitable for studying host immune responses or evaluating immunotherapies. Methods: The virulence of 30 clinical isolates was assessed in mice. The most… Show more

“…In this circumstance, host immune responses exhibit a transitional inflammation in the lung and eventually control the bacterial infection. Recently, more hypervirulent strains of A. baumannii are isolated and used to establish lethal mice model and more researchers explored the governing host factors associated with A. baumannii lethality (9,10). It has been reported that A. baumannii infection induced the production of proinflammatory cytokines TNFα, type I IFN, and IL-1β, which mediated cell death and lung pathology (11)(12)(13)(14).…”

“…The mice were intraperitoneally anaesthetized with pentobarbital sodium (75 mg/kg) and inoculated with 20 μL (2.5×10 8 , 5×10 7 , 1×10 7 , 0 CFU) of A. baumannii by non-invasive intratracheal instillation under direct vision. The survival of the mice was observed for 7 days post infection (8). All animal care and use protocols in this study were performed in accordance with the Regulations for the Administration of Affairs Concerning Experimental Animals approved by the State Council of People’s Republic of China.…”

“…Most laboratory strains and clinical isolates do not cause severe infections in immunocompetent mice, inducing only a self-limiting pneumonia with very limited local bacterial replication and systemic dissemination, even when a large inoculum is used (6, 7). However, studies have shown that some clinical CRAB strains are lethal to immunocompetent mice and thus revealed the range of virulence in different strains of this pathogen (8, 9). LAC-4 is a hypervirulent strain that killed 100% of mice within 48 hours after being inoculated with 10 8 colony-forming units (CFU) (9).…”

“…Specific-pathogen-free grade C57BL/6 6-8 weeks old female mice were purchased from Hunan SJA laboratory animal CO., LTD (Hunan, China). The mice were intraperitoneally anaesthetized with pentobarbital sodium (75 mg/kg), then inoculated with 20 μL (2.5×10 8…”
