A Highly Specific BODIPY-Based Fluorescent Probe for the Detection of Hypochlorous Acid

Abstract: A fluorescent probe, HKOCl-1, has been successfully developed for the detection of hypochlorous acid on the basis of a specific reaction with p-methoxyphenol. The formation of HOCl has been successfully detected not only in an abiotic system but also in an enzymatic system (myeloperoxidase/H2O2/Cl(-) system) and in living macrophage cells upon stimulation. This new probe might be used as an efficient tool for probing the roles HOCl plays in biological systems.

“…6 Functional groups, sensitive to hypochlorite oxidation, such as p-methoxyphenol, 7 dibenzoylhydrazine, 6f thiol, 6d selenide, 6a-c oxime 8 and p-alkoxyaniline 9 have been extensively utilized in the probe design. Some of these probes, however, show pH dependency and function only under basic conditions (pH > 9.0), which is an undesired feature for cell imaging studies.…”

“…Besides APF/HPF, different other fluorescent probes have been tested to specifically detect the halogenating activity of MPO, including sulfonaphthoaminophenyl fluorescein, tetramethylrhodamine, and p-methoxyphenol-coupled boron dipyrromethene (67)(68)(69). However some of these dyes are only partially applicable to flow cytometry and/or are not commercially available.…”
