A fluorescent fatty acid probe, DAUDA, selectively displaces two myristates bound in human serum albumin

Abstract: 11-(Dansylamino) undecanoic acid (DAUDA) is a dansyl-type fluorophore and has widely used as a probe to determine the binding site for human serum albumin (HSA). Here, we reported that structure of HSA-Myristate-DAUDA ternary complex and identified clearly the presence of two DAUDA molecules at fatty acid (FA) binding site 6 and 7 of HSA, thus showing these two sites are weak FA binding sites. This result also show that DAUDA is an appropriate probe for FA site 6 and 7 on HSA as previous studied, but not a goo… Show more

“…The rHSA⅐glucose and rHSA⅐fructose complexes were prepared at a molar ratio of 1:15. The final HSA concentrations in the complexes were about 100 mg/ml, similar to previous studies (22)(23)(24). The complex was then crystallized by sitting drop vapor diffusion at room temperature by mixing 1 l of the protein complex with 1 l of the precipitant solution containing 25-30% (w/v) polyethylene glycol 3350 in 50 mM potassium phosphate at pH 7.5.…”

“…The complete understandings of the unfolding process require a detailed domain specific study and thus establish the complete denaturation landscape. Human serum albumin (HSA, Scheme ) is such a multidomain protein with three different distinguishable domains and has been studied widely in the last few decades . It consists of 585 amino acids residue and is the major protein found in our circulatory system .…”
