The forkhead box transcription factor A2 (FOXA2) is a transcription factor and plays a key role in embryonic development, metabolism homeostasis and tumor cell proliferation; however, its regulatory potential in CRC is not fully understood. Here, it is found that FOXA2 expression is markedly up‐regulated in tumor samples of CRC patients as compared with the normal tissues, which is closely associated with the worse survival in patients with CRC. Notably, a positive correlation between FOXA2 and nuclear factor erythroid 2‐related factor 2 (Nrf2)/glutathione peroxidase 4 (GPX4) gene expression is observed in CRC patients. Mechanistically, FOXA2 depletion weakens the activation of Nrf2 pathway and decreases GPX4 level in CRC cells, thereby leading to ferroptosis, which is further supported by bioinformatic analysis. More intriguingly, the E3 ubiquitin ligase tripartite motif containing 36 (TRIM36) is identified as a key suppressor of FOXA2, and it is observed that TRIM36 can directly interact with FOXA2 and induce its K48‐linked polyubiquitination, resulting in FOXA2 protein degradation in vitro. Taken together, all the studies demonstrate that FOXA2 mediated by TRIM36 promotes CRC progression by inhibiting the Nrf2/GPX4 ferroptosis signaling pathway, thus providing a new therapeutic target for CRC treatment.

The increase in energy consumption and demand is a problem that needs to be resolved immediately, one of the viable solutions is to develop a Dye-Sensitized Solar Cell (DSSC) prototype. This solar cell precursor can also be used as a prototype of STEAM Education for improving students' knowledge, skills, and motivation. Consequently, it is aimed to identify the best preparation technique for DSSC and provide recommendations for designing STEAM Education prototypes in physics learning. This is a preliminary study that employs a quantitative-qualitative descriptive research approach. Based on the results of the output voltage produced by the DSSC, which was given three kinds of immersion variations, quantitative descriptive analysis was carried out to show the type of DSSC with the best preparation technique. Based on the research data, the best preparation technique for DSSC was obtained by immersing the TiO2 layer in a dye solution for 144 hours. There is a positive correlation between the duration of immersion in the dye solution and the voltage generated in the DSSC. Furthermore, the DSSC-based STEAM Education prototype could explain the concept of photo electricity effect, electromagnetic induction, converting light energy into motion, and various other science concepts in applying physics learning. The STEAM Education approach and integrating the three pillars of Education for Sustainable Development explain how science concepts can answer sustainable energy problems related to the environment, society, and economy. This implies preparing students to be the next generation of leaders with the knowledge and abilities to carry on with long-term development plans

Dye-Sensitized Solar Cells (DSSC) are photovoltaic devices that contain a dye that acts as a solar light acceptor. The use of dyesensitized solar cells to solve increasing energy demand and environmental problems still results in low efficiency values. In this study, optimization of DSSC components was carried out to increase DSSC efficiency by varying the thickness of the titanium dioxide (TiO2) semiconductor photoanode layer, polyvinylidene fluoride (PVDF) trap electrolyte membrane, and polyvinylidene fluoride nanofiber (PVDF NF) to obtain the optimum thickness. Scanning Electron Microscope (SEM) results of membrane thickness variation and titanium dioxide (TiO2) semiconductor photoanode coating showed the formation of nanofiber fibers composed of three-dimensional, porous, and diameter networks connected to the PVDF NF membrane. The increase in density and decrease in pore size, along with an increase in thickness and cracking as the TiO2 photoanode semiconductor layer increases, affect the electron transport rate of the DSSC. The higher particle density level will inhibit the electron transport rate, so it can reduce the efficiency of DSSC. The optimum thickness of the TiO2 semiconductor layer and PVDF NF electrolyte membrane of 0.20 mm and 0.35 mm can produce values, voltage, fill factor current density, and electrical efficiency of 500 mV, 2.7 x 10−3 mA.cm−2, 1.80%, and 2.40%, respectively.
