A concentration-dependent endocytic trap and sink mechanism converts Bmper from an activator to an inhibitor of Bmp signaling

Abstract: Bmper, which is orthologous to Drosophila melanogaster crossveinless 2, is a secreted factor that regulates Bmp activity in a tissue- and stage-dependent manner. Both pro- and anti-Bmp activities have been postulated for Bmper, although the molecular mechanisms through which Bmper affects Bmp signaling are unclear. In this paper, we demonstrate that as molar concentrations of Bmper exceed Bmp4, Bmper dynamically switches from an activator to an inhibitor of Bmp4 signaling. Inhibition of Bmp4 through a novel en… Show more

“…HSPGs can also bind to BMP7 and either promote or inhibit signaling (45)(46)(47). Other ECM components or ECM-bound proteins, such as fibronectin, collagen, and members of the crossveinless family, promote BMP signaling through diverse and context-dependent mechanisms (48)(49)(50)(51)(52). The composition of the ECM varies between different tissues and cell types and thus it is feasible that the BMP4 prodomain might be required to facilitate interactions with obligate ECM binding partners present on ectodermal, but not mesodermal cells.…”

“…sfrp5 has been described as a Wnt inhibitor in several systems (27,28), an integrator of Wnt-BMP signaling in the zebrafish gut (29), and a regulator of mouse incisor renewal (30). bmper has been reported as both a positive and negative mediator of BMP signaling in other organs of the mouse, frog, and fly (31)(32)(33), and as an antagonist of BMP in mouse incisor ameloblasts (34). We assayed expression of these candidates in species with divergent densities of teeth/taste buds: C. afra, a species with few teeth and taste buds and Labeotropheus fueleborni, a species with many of both (SI Appendix, Fig.…”

“…In zebrafish, chordin becomes a significantly more effective BMP inhibitor in vivo following cleavage of the C-terminal domain (16). However, there is significant evidence that chordin fragments have reduced biological activity in Xenopus (1,17) and are subject to more rapid endocytotic turnover (18). Chordin interacts with cell surface proteins including crossveinless-2 (CV2) (19) and collagen IV (20).…”
