A (1→3,1→4)‐β‐glucan‐specific monoclonal antibody and its use in the quantitation and immunocytochemical location of (1→3,1→4)‐β‐glucans

Abstract: SummaryMonoclonal antibodies were raised against a (1-~3,1~4)-~-glucan-bovine serum albumin (BSA) conjugate. One antibody (BG1) selected for further characterization, was specific for (1--~3,1--~4)-~-giucan, displaying no binding activity against a (l~3)-~-glucan-BSA conjugate and minimal binding against a cellopentaose-BSA conjugate. A range of oligosaccharides was prepared by enzymatic digestion of (1-~3,1-~4)-13-glucan, purified by size exclusion chromatography and characterized by 1H-NMR and anion exchange… Show more

“…The principal differences were the enhanced amounts of glucomannans in epidermal cell walls, and an enhancement of the ␤-glucan in the mesophyll cell walls, about 4-fold higher than in epidermal walls (Table II). Meikle et al (1994) demonstrated the specificity of the monoclonal antibody BG1 against cereal mixed-linkage ␤-glucans, and found this epitope to be distributed uniformly across the aleurone walls of wheat (Triticum aestivum) endosperm cells but excluded from the middle lamella. The ␤-glucan is distributed uniformly across the thin walls of the mesophyll cells, whereas it is concentrated primarily on the innermost portion of the outer facing wall of the epidermis (Fig.…”

“…Grain samples from 10 to 28 DAP were prepared for microscopy and changes to the endosperm were monitored by taking transverse sections of the developing grains. The distribution and relative abundance of polysaccharides in the differentiating grain were detected on sections using monoclonal antibodies raised against specific polysaccharide epitopes including (1→3)-b-D-glucan (Meikle et al, 1991), xyloglucan LM15 (Marcus et al, 2008), (1→3,1→4)-b-D-glucan (Meikle et al, 1994), hetero-(1→4)-b-Dmannan (Pettolino et al, 2001), and arabino-(1-4)-b-D-xylan LM11 (McCartney et al, 2005). Sections for light microscopy (1 mm) were either stained in toluidine blue to highlight tissue architecture or processed for immunogold labeling.…”

“…Thin sections (100 nm) were cut on an ultramicrotome (Leica) and picked up on carbon-coated and plastic-filmed gold grids. The (1/3),(1/4)-b-D-glucan epitopes were detected with the BG1 monoclonal antibody that specifically recognizes this mixed-linkage b-glucan (Meikle et al, 1994; available from Biosupplies Australia), and visualized by a 10-nm colloidal gold-conjugated rabbit anti-mouse secondary antibody (Sigma). The blocking solution was 1% (w/v) acetylated bovine serum albumen (Aurion) in phosphate-buffered saline plus 0.1% (w/v) Tween 20, and incubation solution for washes was 0.1% acetylated bovine serum albumen in phosphate-buffered saline plus 0.01% (w/v) Tween 20.…”

“…The BG1 monoclonal antibody recognizes the (1/3),(1/4)-b-D-glucan with high specificity (Meikle et al, 1994). This monoclonal antibody has been used to show dramatic changes in epitope abundance of (1/3),(1/4)-b-D-glucan in the cell walls of developing tissues (Meikle et al, 1994;Trethewey et al, 2005;McCann et al, 2007) and its appearance in the cell walls of Arabidopsis (Arabidopsis thaliana) following heterologous expression of genes thought to encode its synthases Doblin et al, 2009).…”
