Abstract: The adenosine A1 receptor selective agonist, N6-cyclopentyladenosine (CPA, 300 nM) inhibited basal accumulation of [3H]inositol phosphates ([3H]InsPs), but not the total levels of membrane [3H]-phosphoinositides, in rat hippocampal slices. This action of CPA was not significantly modified when synaptic transmission was blocked with tetrodotoxin (TTX, 200 nM) but was prevented in slices pre-incubated with pertussis toxin (PTX, 5 microg/mL) for 12-16 hr. Neither PTX nor TTX, when applied in the absence of CPA, i… Show more

“…Yet, in mouse cerebral cortical slices [92] stimulation of A 1 Rs resulted in decreased histamine-induced IP 3 formation. Additionally, in rat hippocampal slices selective A 1 R agonists inhibit PLC basal activity through G i/o proteins [93,94]. As will be discussed in the next sections, either an increase or decrease in PLC activity could mediate the inhibitory effects of A 1 R activation.…”

“…It is coupled to G i/o proteins, inhibiting adenylyl cyclase and consequently decreasing cyclic adenosine monophosphate (cAMP) formation and protein kinase A (PKA) activation ( Fredholm et al, 2001 ; Serpa et al, 2015a ). Furthermore adenosine A 1 receptor also regulates phospholipase C activity ( Cascalheira and Sebastião, 1998 ; Cascalheira et al, 2002 ), inhibits N- and P/Q-type calcium channels ( Ambrósio et al, 1997 ) and activates inwardly rectifying potassium channels ( Takigawa and Alzheimer, 2002 ).…”
