5 nm Gold-Ni-NTA Binds His Tags

Abstract: Molecular biology has benefited by tags encoded into expressed proteins for their identification and purification. The protein of interest is constructed with DNA that includes an added "tail" sequence coding for a string of amino acids that bind to an antibody, a metal column, or a some other binding couple. Common genetic tags include 6x-histidines that bind Ni-NTA, Flag that binds to anti-flag antibodies, strep-tag that binds to strep-tactin, c-myc that binds to antibody, calmodulin binding domain that bind… Show more

“…In this regard we have labeled the His-tagged ISWI with Ni-NTA 5-nm gold cluster and it can next be used to mark this protein in the complex (Reddy et al, 2005). Another approach is to dock X-ray crystallography structure of subunits into the EM 3D density map.…”

“…Since the C-terminus of TRIM5α CC-SPRY is tagged with His 6 , we used Ni-NTA nano-gold labeling to confirm its binding to CA assemblies (52). The method is described in detail here.…”

“…To further investigate how CC1∆C223 binds microtubules, we co-polymerized tubulin in the presence of CC1ΔC223. We then labeled CC1ΔC223 using 5 nm gold-conjugates that recognize the His-tag 21 and monitored the formed microtubules and gold distribution via transmission electron microscopy (TEM). Gold labeling only occurred at closely aligned microtubules with very small inter-microtubule distances (Fig.…”
