“…We prepared biotinylated mouse anti-HA IgG1 (31), parental anti-RP105 mAb (rat), mouse anti-MD-1 (JR7G1) (35), and unconjugated anti-HA IgD (31). The parental anti-RP105 mAb (rat) was obtained from the supernatant of the hybridoma (14) ( Figures 1B, C) or purified from ascites fluid, which was obtained from pristane (Funakoshi, Tokyo, Japan)-primed nude mice that were intraperitoneally inoculated with the hybridoma (36,37) using the caprylic acid (Wako, Tokyo, Japan)-ammonium sulfate (Katayama Kagaku, Osaka, Japan) precipitation method (38) (Figures 2A, B). The concentrations of the parental anti-RP105 mAb (rat) and aRP105 were determined by quantitative ELISA using goat anti-mouse Ig (Southern Biotech) and HRP-conjugated goat anti-mouse IgG (Southern Biotech) as described in the ELISA section.…”

“…Immunosuppre ssed mice, rats, (132) and hamsters (1 33) have also been used to grow interspecific hybridomas. X-radiation or drug-induced immunosuppre ssion can be used.…”

“…Although murine monoclonal antibodies can be produced in high concentrations by growth of cells as ascitic tumors in histocompatible mice, similar attempts to grow heterohybridomas and xenogenic hybridomas have been much less successful (15,16,17,18,19,20). Methods such as X-ray irradiation of mice (16,17); treatment with immunosuppressive drugs such as antilymphocyte globulin, methotrexate, cyclophosphamide (16) or hydrocortisone (17); prior subcutaneous adaptation of hybridoma cells (15); and the use of T-cell deficient (15,16,18) or T-cell and B-cell deficient SCID mice (19) have been employed in attempts to improve production.…”

Even when utilizing one or more than one of these methods simultaneously, the efficiency of take for human hybridomas in mice may be as low as 50% (20).…”
