Photoresponsive Formation of an Intermolecular Minimal G‐Quadruplex Motif

Abstract: The ability of three different bifunctional azobenzene linkers to enable the photoreversible formation of a defined intermolecular two-tetrad G-quadruplex upon UV/Vis irradiation was investigated. Circular dichroism and NMR spectroscopic data showed the formation of G-quadruplexes with K(+)  ions at room temperature in all three cases with the corresponding azobenzene linker in an E conformation. However, only the para-para-substituted azobenzene derivative enables photoswitching between a nonpolymorphic, stac… Show more

“…[144,145] Realizing that ATP (adenosine triphosphate) [146,147] and VEGF (vascular epithelial growth factor) [148] are over-expressed in cancer cells, functional all-DNA microcapsules responding to these biomarkers were developed (Figure 7A). In this system, the layer-by-layer hybridization of the DNA scaffolds composed of two units of (28) and (29) and two units of (30) and (29) was used to construct the shells. For the ATP-responsive microcapsules, two DNA scaffolds composed of two strands of (25) hybridized with (26) and of two strands of (27) hybridized with (26) were used to construct the DNA shell by the layer-by-layer hybridization process, where the bridging units (26) included the specific anti-ATP aptamer sequence in a "caged" configuration.…”

“…CaCO 3 microparticles impregnated with the respective substrates were coated with the basic, positively charged PAH layer. The interlayer bridging units (29) included the specific anti-VEGF aptamer sequence. Similarly, by coating the PAH-modified CaCO 3 particles, the VEGF-responsive microcapsule shells were constructed.…”

“…The non-nucleotide spacers has been shown to increase the thermal stability of the structure of duplexes, triplexes or tetraplexes (2329). They were also implemented in nuclear magnetic resonance (NMR) (3034) and crystallographic studies (29,35,36) but so far such linkers has not been used to stabilize the native structure of RNA hairpins.…”
