The aim of this study was to prepare 3-hydroximino- and 3-amino derivatives of glycyrrhetinic acid and derivatives to evaluate their in vitro cytotoxicity for a panel of human tumor cell lines. Thus, commercially available glycyrrhetinic acid (1) was acetylated or oxidized at position C-3 and transformed into a variety of different esters and amides followed by their conversion to 3-oximes and amines. While the parent compound was not cytotoxic at all, the 3-amino esters are highly cytotoxic. Interestingly, 3-amino amides were significantly less cytotoxic than 3-amino esters. The (3b, 18b, 20b) Benzyl 3-amino-11-oxoolean-12-en-30-oate was the most cytotoxic compound of this series showing an EC50 = 1.3 mM for 518A2 melanoma cells.

Beta-boswellic acids are triterpenoids being generic to the plants of genus boswellia. Although they were shown to exhibit different biological activities, the cytotoxic potential of b-boswellic acid derivatives remained by and large unexploited. To expand the potential of these compounds we developed simple procedures for the interconversion of the most important b-boswellic acids 1-4 and prepared several other derivatives 5-48. These compounds were screened for their cytotoxic activity in sulforhodamine B assays employing several human tumor cell lines and nonmalignant mouse fibroblasts. One of these compounds, a difluoromethylester of 3-O-acetyl-11-keto-beta-boswellic acid 23, was cytotoxic for human breast adenocarcinoma cells MCF-7 (EC50 = 6.5 mM) while being significantly less cytotoxic for the mouse fibroblasts.

ZusammenfassungHarze der Bäume verschiedener Boswellia-Spezies – pharmazeutisch Olibanum genannt – werden seit Jahrtausenden als Arzneimittel – auch bei Tumoren – verwendet. Seit den 1990er-Jahren häufen sich die Berichte über Antitumorwirkungen von Extrakten aus dem Harz und einigen Inhaltsstoffen, hier insbesondere von Boswelliasäuren. Präklinische Studien zeigten eine Hemmung der Proliferation, Antiangiogenese und der Ausbreitung von Metastasen sowie Induktion von Apoptose bei einer Vielzahl von Tumorzelllinien wie: Leukämie-, Glioblastom-, Meningiom-, Prostata-, Leber-, Mamma-, Kolon-, Pankreas- und Lungenkarzinomzellen.Die Inhaltsstoffe eines Boswelia-Extraktes (BE) verhalten sich in ihrer Antitumorwirkung synergistisch zueinander und auch gegenüber Chemotherapeutika und Bestrahlung. Der Mechanismus der Antitumorwirkung beeinflusst mehrere Transduktionssignalwege und Einzelfaktoren, die mit Proliferation, Apoptose, Angiogenese und Invasion im Zusammenhang stehen. Ihre Aktivierung erfolgt in vielen Fällen durch Phosphorylierungsreaktionen. Letztere werden durch BE und -inhaltsstoffe gehemmt. Eine wichtige Rolle bei der Apoptosewirkung spielt die Aktivierung von Caspasen.Klinische Studien liegen außer bei Hirntumoren nicht vor. Ein Vorteil von BE ist die geringe Toxizität. Dies sollte Anlass zur Durchführung klinischer Studien sein.
