Abstract: OBJECTIVES:The promotion of extracellular matrix synthesis by chondrocytes is a requisite part of an effective cartilage tissue engineering strategy. The aim of this in vitro study was to determine the effect of bi-axial cyclic mechanical loading on cell proliferation and the synthesis of glycosaminoglycans by chondrocytes in three-dimensional cultures.METHOD:A strain comprising 10% direct compression and 1% compressive shear was applied to bovine chondrocytes seeded in an agarose gel during two 12-hour condit… Show more

“…The full thickness of the cartilage tissue from the entire proximal surface of the joint was removed under sterile conditions. Cartilage explants were enzymatically digested with protease and collagenase (both from SIGMA‐ALDRICH (St. Louis, MO) . Briefly, 20 U mL −1 protease was applied for 1 h at 37°C and further immersed in 100 U mL −1 of collagenase type II for further 16 h at 37°C.…”

“…Cartilage explants were enzymatically digested with protease and collagenase (both from SIGMA-ALDRICH (St. Louis, MO). 35 Briefly, 20 U mL 21 protease was applied for 1 h at 378C and further immersed in 100 U mL 21 of collagenase type II for further 16 h at 378C. After the digestion period, cell, and collagenase suspension was passed through 70 mm cell sieve.…”

“…A similar approach shown in figure 3b studied how fibroblasts change their orientation according to the direction of the stretching [90]. In addition to stretching, compression is also an important stimulation for simulating the environment for cells forming bones and cartilage [91][92][93][94]. Also, stem cells have been found to differentiate on a chip under straining stimulation simulating the vascular system [95], as illustrated in figure 3b.…”

“…These applied forces simulate loading that would be normally exerted during activities of daily movement. Specifically, cyclical loading of articular chondrocytes has been shown to increase extracellular matrix production and cell viability within engineered cartilage constructs [101,102]. Other bioreactors have been developed to provide dynamic changes in hydrostatic pressure, tension, shear, vibrations, etc.…”
