Mutagenesis of Puccinia graminis f. sp. tritici and Selection of Gain-of-Virulence Mutants

Abstract: Wheat stem rust caused by the fungus Puccinia graminis f. sp. tritici (Pgt), is regaining prominence due to the recent emergence of virulent isolates and epidemics in Africa, Europe and Central Asia. The development and deployment of wheat cultivars with multiple stem rust resistance (Sr) genes stacked together will provide durable resistance. However, certain disease resistance genes can suppress each other or fail in particular genetic backgrounds. Therefore, the function of each Sr gene must be confirmed af… Show more

“…Primary (T 0 ) hemizygous transgenic plants and non-transgenic controls, which had also undergone tissue culture, were phenotyped with Puccinia graminis f. sp. tritici isolate UK-01 as previously described 68 , and the infection phenotypes were correlated with the presence of the hpt transgene (Fig. 6c; Supplementary Table 14).…”

“…There are several approaches to cloning effector genes. Variation can be generated through mutagenesis (Salcedo et al, 2017;Kangara et al, 2020), but there are challenges associated with using mutant populations to clone effector genes: the presence of more than one nucleus in some pathogen species (e.g., dikaryotic Puccinia spp. and stinking smut, Tilletia tritici), laborintensive bulking, applying an appropriate mutagen dose, and the difficulty of accurately identifying mutant variants.…”
