Abstract: Psoriasis is a chronic inflammatory skin disorder characterized by hyperproliferative keratinocytes and immune cell infiltration into the skin, often accompanied by itch. Histamine, acting via histamine 1–4 receptors, is known to modulate immune responses in the skin and to induce itch. The aim of this study was to test the role of histamine 2 receptors and histamine 4 receptors in the imiquimod-induced psoriasis-like skin inflammation model. BALB/c mice were treated intraperitoneally with amthamine (histamine… Show more

“…Interestingly, H4R blockade improved systolic function similarly to H1R and H2R blockade, and improved mortality, as did H2R blockade. These are interesting findings since the H4R has been shown to promote inflammatory cell infiltration and cytokine production in mouse models of lung fibrosis [156], psoriasis [157], and colitis [158]. With the critical contribution of inflammation to adverse myocardial remodeling, there appears to be a basis for the H4R contributing to this process, although clearly more studies are needed to establish this.…”

“…Mommert et al [28] found that stimulation of H4 receptor, which is highly expressed on plasmocytoid dendritic cells (pDC) in psoriasis [29], increases production of interleukin 17 (IL-17), a cytokine that plays a major role in the pathogenesis of psoriasis. Recently, it has been also shown that blockade of H4R may help to ameliorate imiquimod-induced skin inflammation, diminish epidermal hyperproliferation, and inhibit spontaneous scratching behavior in mice [30]. These observations suggest that histamine relevance in the pathophysiology of pruritus in psoriasis is still uncovered and further investigations are needed.…”

“…Imiquimod (IMQ) is an immune response modifier clinically used to treat genital and perianal warts, actinic keratosis, and superficial basal cell carcinoma [ 1 ]; this compound is also experimentally used to induce psoriasis-like skin inflammation in mouse models [ 2 , 3 , 4 ], and as an adjuvant to weakly immunogenic vaccines [ 5 , 6 , 7 ]. Despite being a small molecule (molar mass 240 g/mol), IMQ has low aqueous solubility and unsatisfactory cutaneous permeability, limiting its formulation into effective dosage forms [ 8 ].…”
