Mendel's Stem Length Gene (Le) Encodes a Gibberellin 3b-Hydroxylase

Abstract: We describe the isolation of the Le gene of pea, which controls internode elongation and originally was described by Mendel. Heterologous screening of a pea cDNA library yielded a partial clone that was 61% identical to coding regions of the putative Arabidopsis gibberellin 3 beta-hydroxylase gene, GA4. DNA gel blot analysis with this cDNA revealed a HindIII restriction fragment length polymorphism between pea isolines differing at Mendel's Le locus. Genomic clones of the GA4-related gene were isolated from th… Show more

“…Another key GA gene from pea is Mendel's LE, also known as the 3-oxidase gene PsGA3ox1 (Lester et al, 1997;Martin et al, 1997). Mendel exploited the dwarf stature of mutant le-1 shoots in his original genetics experiments, but it is interesting to note that the roots of le-1 plants (and of other le mutants) are indistinguishable from the wild type, and contain normal GA levels (Yaxley et al, 2001).…”

“…The expression levels of the 3-oxidase genes PsGA3ox1 (also known as Mendel's LE; Lester et al, 1997;Martin et al, 1997) and PsGA3ox2 were measured in the shoot and root tissue of 6-d-old pea seedlings using real-time PCR. PsGA3ox1 was more highly expressed in the shoot compared with root tissue, with an approximate 2-fold difference (P , 0.001; Fig.…”

“…LE was the second of Mendel's genes to be cloned when in 1997 two groups working independently reported the successful isolation of this gene (Lester et al 1997;Martin et al 1997). This was made possible by the previous isolation (by transposon tagging) of a putative GA 3b-hydroxylase gene from Arabidopsis, GA4 (Chiang et al 1995).…”

“…Seed shape (smooth versus wrinkled) and stem length are regulated by the R and Le loci and correspond to natural mutations of a starch-branching enzyme gene (13) and a gibberellin 3␤-hydroxylase gene (14,15), respectively. Cotyledon color (yellow versus green) is regulated by the I locus (16), with the i mutant retaining not only greenness of the cotyledon during seed maturation but also greenness of leaves during senescence, suggesting that this mutant is a stay-green mutant (17).…”
