Abstract: Purpose To investigate the role and related mechanisms of miR-106a in sepsis-induced AKI. Methods Serum from sepsis and healthy patients was collected, sepsis mouse model was established by cecal ligation and puncture (CLP). TCMK-1 cells were treated with lipopolysaccharide (LPS) and transfected with THBS2-small interfering RNA (siTHBS2), miR-106a inhibitor, miR-106a mimics and their negative controls (NCs). The expression of miR-106a, thrombospondin 2 (THBS2), Bax, cle… Show more

“…The anti-proliferative and anti-angiogenic effects of PGA were confirmed here by the reduction of pro-inflammatory corneal cytokine levels (Il-1α, Il-1β, Il-6 and TNF-α), paralleled by an increment in anti-inflammatory cytokines (Il-10, Il-1rα). Particularly, cytokine levels were modulated by miRNAs strictly involved in LPS response [ 62 , 65 , 66 , 67 ] and known to modulate TLR4-dependent cytokines, such as TNF-α and Il-β, targeted by both miR-20a-5p and miR-106a-5p [ 53 , 68 ]; Il-10, regulated by miR-27a-5p [ 67 ], and Il-6, modulated by miR-106a-5p [ 53 ] Overall, PGA was able to increase miR-20a-5p and miR-106a-5p, which were reduced by LPS intra-stromal injection. The increase of these miRNA by PGA was paralleled by the reduction of three important pro-inflammatory cytokines such as Il-6, TNF-α and Il-1β.…”

“…The mitochondrion is assumed to be a key organelle in the development of septic acute kidney injury, and has major pathophysiological significance in ROS production and apoptosis [ 27 ]. In one hybrid human and experimental study including 50 patients with sepsis, an effect was found of miR-106a on caspase-3 activity, BCl-2 expression and proinflammatory cytokine production after LPS stimulation [ 28 ]. The authors found an association between miR-106a and an aggravation of LPS-induced inflammation, and apoptosis in sepsis-induced AKI.…”

“…The authors found an association between miR-106a and an aggravation of LPS-induced inflammation, and apoptosis in sepsis-induced AKI. A target gene for miR-106a was established as thrombospondin THBS2 , which takes part in a number of processes such as regulation of cell motility, death and cytoskeleton formation [ 28 ].…”

“…Recent studies demonstrated microRNAs (miRNAs/miRs), as endogenous non-coding RNA molecules are important mediators in degrading mRNA or inhibiting translation of their target genes to act as crucial molecular markers in diagnosis and prognosis of acute kidney injury [17,18]. By searching for related studies, we found that most of studies focused on the role of miRNAs in ischemic acute kidney injury, while its role in sepsis-induced acute kidney injury is limited [19], including miR-21 [20], miR-204 [21], miR-290-5p [22] and miR-106a [23]. Interestingly, a recent study by Ge et al [24] reported the markedly decreased miR-22-3p expression in sepsis-induced kidney injury.…”
