Inhibitory effect of Astragalus polysaccharide on osteoporosis in ovariectomized rats by regulating FoxO3a /Wnt signaling pathway

Abstract: Purpose: To investigate inhibitory effect of Astragalus polysaccharide (APS) on osteoporosis in ovariectomized rats by regulating FoxO3a/Wnt2 signaling pathway. Methods: Postmenopausal osteoporosis (PMOP) animal model was developed by excising the bilateral ovaries of rats. The model rats were administered with APS (200 mg/kg, 400 mg/kg, 800 mg/kg) by intragastric administration once daily for 12 weeks. Bone density, bone metabolism index and oxidative stress index were… Show more

“…Other studies have shown that APS had signi cant antitumor activity in A549 and NCI-H358 human non-small cell lung cancer cells [28]. Ou et al established a mouse model to con rm that APS could promote bone formation by activating the Wnt/LRP5/β-catenin pathway [29]. The preliminary results of this study con rmed that APS could inhibit the differentiation and maturation of OCs and thus disrupt the balance between OBs and OCs, inhibit the proliferation, invasion, and metastasis of lung adenocarcinoma cells, and nally inhibit lung adenocarcinoma bone metastases.…”

“…For example, previous studies confirmed that Epimedium and its components could antagonize bone resorption and stimulate bone formation [35] and had a positive effect on the reconstruction of bone biological function [36]. Astragalus polysaccharide, the active substance of astragalus, has been proven to effectively inhibit osteoclastogenesis and increase bone mass [37,38] and prevent osteoporosis in ovariectomized mice [39]. Some studies have found that Eucommia can effectively inhibit the reduction of bone density caused by OVX, improve bone strength, effectively improve the obesity caused by estrogen reduction, and have no hyperplastic effect on the uterus [40,41].…”

“…The administration of Astragalus polysaccharide in male Sprague-Dawley rats enhanced bone marrow mesenchymal stem cell (BMSC) differentiation and proliferation by triggering Wnt/β-catenin and PI3K/AKT pathways through bone morphogenetic protein type 9 (BMP9) overexpression and miR-152 down-regulation (Li et al 2019a, b). Daily administration of Astragalus polysaccharide, in ovariectomized rats, for 3 months, alleviated the osteoporosis caused by oxidative stress via the regulation of FoxO3a/Wnt2/β-catenin pathway (Ou et al 2019). Astragaloside-IV extract administration to rats with ischemia-reperfusion injury showed neuroprotective activity, regulated the expression of apoptosis-related genes, improved the pathological brain damage and declined the cerebral infarction volume (Yin et al 2020).…”
