Abstract: Since its discovery, myostatin (MSTN) has been at the forefront of muscle therapy research because intrinsic mutations or inhibition of this protein, by either pharmacological or genetic means, result in muscle hypertrophy and hyperplasia. In addition to muscle growth, MSTN inhibition potentially disturbs connective tissue, leads to strength modulation, facilitates myoblast transplantation, promotes tissue regeneration, induces adipose tissue thermogenesis and increases muscle oxidative phenotype. It is also k… Show more

“…This finding may be explained by the physiological limit of the tissue or animal, which takes into account the tissue adaptation limit regulated by myostatin, thus preventing hypertrophy occurring in an uncontrolled manner (Yamada et al, 2012).…”

“…Moreover, future HDTBR campaigns should also focus on simulating upcoming Lunar Gateway mission profiles where the crew will transition from prolonged exposure to microgravity to hypogravity on the Lunar surface. Consequently, 'conventional' exercise stimuli such as high reaction forces and high muscle forces to stimulate bone growth (Frost, 2003), and high load resistive exercises to promote muscle hypertrophy (Yamada et al, 2012)-as is currently the case-will likely become of secondary importance. For Lunar Gateway missions with Lunar surface EVAs after a prolonged exposure to microgravity, the primary needs and requirements of the countermeasure programmes may probably undergo a shift from focussing on maintaining bone mineral density, muscle strength and VO2max to countermeasures focussing on orthostatic tolerance, postural stability, spatial orientation and balance due to the transition between microgravity and hypogravity, ultimately to assure crew safety and mission success.…”

“…Moreover, the same interventions that act preventively in at-risk patients, such as a muscular dystrophy gene-editing protocol to preserve normal strength and endurance, might enhance the target traits beyond the normal range when applied to healthy people. 20 Policing such ''off-label'' uses of these preventive interventions is likely to go beyond the jurisdiction of gene-editing research governance to require responses by public regulatory bodies such as the U.S. Food and Drug Administration. However, gathering the safety and efficacy evidence that such responses will require will mean conducting research on the enhancement potential of these preventive interventions.…”
