Neosporosis is an emergent disease responsible for considerable economic impact due to reproductive losses. Its zoonotic potential remains unknown. This study involved a survey of antibodies to Neospora caninum in slaughtered sheep and their association with epidemiological variables. Serum samples from 596 sheep from the states of São Paulo and Rio Grande do Sul, Brazil, were collected in two slaughterhouses located in São Paulo and evaluated by indirect fluorescence antibody test (IFAT), using cut-off titers of 25. Among these samples, 353/596 (59.23%; 95%CI 55.23-63.10) were positive and 263/353 (74.50%; 95%CI 69.71-78.77%) were from Rio Grande do Sul. Statistical associations were determined in the univariate analysis between the serological results and sex, breed and municipality of origin. Sheep that came from extensive breeding system showed higher chance (OR=2.09) of presenting antibodies to N. caninum in relation to those from semi-intensive system. Higher chance was also observed for the different studied breeds, except Bergamácia, in relation to Hampshire Down. The results revealed the presence of infection by N. caninum in sheep from slaughterhouses.Keywords: Neospora caninum, serology, sheep, slaughterhouses. ResumoA neosporose é uma doença emergente responsável por considerável impacto econômico devido a perdas reprodutivas e seu potencial zoonótico permanece desconhecido. Este estudo envolveu a pesquisa de anticorpos para Neospora caninum em ovinos abatidos e sua associação com variáveis epidemiológicas. Amostras de soro sanguíneo de 596 ovinos, procedentes dos estados de São Paulo e Rio Grande do Sul, Brasil, foram colhidas em dois abatedouros localizados em São Paulo, e avaliadas pelo teste de reação de imunofluorescência indireta (RIFI), utilizando-se como ponto de corte o título de 25. Dentre essas amostras, 353/596 (59,23%; IC95% 55,23-63,10) foram positivas e dentre os ovinos positivos 263/353 (74,50%; IC95% 69,[71][72][73][74][75][76][77][78]77%) eram procedentes do Rio Grande do Sul. Associações estatísticas foram determinadas na análise univariada entre os resultados sorológicos e o sexo, raça e município de origem. Ovinos oriundos de sistema de criação extensivo demonstraram maior chance (OR=2.09) de apresentarem anticorpos para N. caninum em relação aos de sistema semi-intensivo. Uma maior chance também foi observada para as diferentes raças estudadas, exceto Bergamácia, em relação à raça Hampshire Down. Os resultados revelaram a presença de infecção por N. caninum em ovinos de abatedouros.

The aim of this study was to determine the occurrence of antibodies IgG against Neospora caninum and evaluate the risk factors associated with the infection in ovine herds, State of Sergipe, Brazil. Blood samples (n=1200) were collected from sheep raised in 60 sheep run located in the three mesoregions of the State of Sergipe, between 2011 e 2012. Antibodies were investigated by Indirect Immunofluorescence Antibody Test of which cutoff point was 50 and positive samples were diluted in base 2 until the last positive titer. Data from 15 variables was obtained from questionnaires given to farmers. Absolute and relative frequencies were determined and the risk factors were analyzed by Pearson's Qui-Square test (p≤0,05). The occurrences of serum reactive sheep were 39.83% (478/1200). The occurrences of positive sheep and sheep run were 55.88% (380/680) and 88.24% (30/34) in the Eastern region; 21.42% (60/280) e 42.85% (6/14) in dry region and 15.83% (38/240) e 41.67% (5/12) in the backwoods respectively. Antibody titers ranged from 50 (n=459), represented 96.02% (459/478) of seropositive samples to 6400 (1/478). Among the significant variables in the multivariate analysis were considered risk factors for infection with N. caninum were, sheep run located in Eastern region (p=0.000, OR=4.64, CI95%=3.36-6.41); standing and running water sources (p=0.000 OR=2.03, CI95%=1.41-2.92), absence of quarantine (p=0.000, OR=2.71, CI95%=2.08-3.53), absence of dunghill (p=0.000, OR=3.14, CI95%=2.45-4.02), presence of dogs (p=0.000, OR=2.74, CI95%=1.73-433), presence of wild animals (p=0.000, OR=3.45, CI95%=2.44-4.87) and subsistence (p=0.000, OR=4.99, CI95%=3.15-7.92) or reproduction (p=0.002, OR=1.74, CI95%=1.22-2.49) livestock were important risk factors. Our results highlight the occurrence of N. caninum in the ovine herds from State of Sergipe. Neospora caninum is protozoan parasite with domestic and wild dogs, coyotes and grey wolves as the definitive hosts and many warm-blooded animals as intermediate hosts. It was cultivated and named in 1988. Neosporosis is a major disease of cattle and has no public health significance. Since 1990's N. caninum has emerged as a major cause of abortion in cattle worldwide, including in Brazil. N. caninum also causes clinical infections in several other animal species. Considerable progress has been made in understanding the biology of N. caninum and there are more than 200 papers on this subject from Brazil. However, most of the reports on neosporosis from Brazil are serological surveys. Overall, little is known of clinical neosporosis in Brazil, particularly cattle. The few reports pertain to sporadic cases of abortion with no information on epidemics or storms of abortion. The objective of the present review is to summarize all reports from Brazil and suggest topic for further research, including prevalence of N. caninum oocysts in soil or in canine feces, and determining if there are additional definitive hosts, other than the domestic dog. There is need for a national survey in cattle using defined parameters. Future researches should focus on molecular characterization of N. caninum strains, possibility of vaccine production and relationship between wildlife and livestock epidemiology.Keywords: Neospora caninum, neosporosis, domestic animals, wild animals, Brazil. ResumoNeospora caninum é um protozoário parasita que possui os canídeos domésticos e selvagens, coiotes e lobos cinzentos como hospedeiros definitivos e vários animais de sangue quente como hospedeiros intermediários. Foi cultivado e nomeado em 1988. A neosporose é uma das principais doenças em bovinos e não tem significância em saúde pública. Desde 1990, N. caninum tem emergido como uma das principais causas de aborto em bovinos em todo o mundo, inclusive no Brasil. N. caninum também causa infecções clínicas em várias outras espécies animais. Consideráveis avanços foram feitos na compreensão da biologia desse parasita e há mais de 200 trabalhos sobre o assunto no Brasil. No entanto, a maioria dos relatos de neosporose do Brasil são relacionados a sorologia. Em geral, pouco se sabe sobre a neosporose clínica no Brasil, particularmente em bovinos. Os poucos relatos referem-se a casos esporádicos de aborto sem informações sobre epidemias ou surtos de aborto. O objetivo da presente revisão é resumir todos os relatos sobre N. caninum no Brasil e sugerir tópicos para pesquisas futuras, incluindo a prevalência de oocistos de N. caninum no solo ou em fezes caninas e determinar se há hospedeiros definitivos adicionais, exceto o cão doméstico no país. Uma pesquisa nacional em bovinos usando parâmetros definidos seria de grande importância.
