Hematological disorders detected in dogs infected by ABSTRACTA retrospective review of hematological reports of nine dogs detected with Hepatozoon canis infection by microscopic examination of blood smears in a laboratory in the municipality of Dourados, Mato Grosso do Sul, Brazil was conducted. This study aimed to evaluate the hematological profile of these infected dogs, in addition to the occurrence of coinfections with other agents that infect blood cells, since studies concerning canine hepatozoonosis in Brazil are scarce and there are some divergences regarding H. canis infection that still require a resolution. The nine cases of H. canis infection were identified among all dogs examined at the studied laboratory in 2009 and 2010, with an occurrence of 7/1,192 (0.59%; 95% CI 0.15 -1.02%) positive dogs in the first year and 2/1,313 (0.15%; 95% CI 0.02 -0.55%) cases in 2010. The analysis of the hematological reports showed an occurrence of coinfection between H. canis and other agents in two (2/9; 22.22%; 95% CI 2.81 -60.00%) dogs, one with E. canis and another with Babesia spp. (1/9; 11.11%; 95% CI 0.28 -48.24%). Only the blood test of one dog had no alterations, based on reference values. Anemia was the most frequent hematological alteration (6/9; 66.67%; 95% CI 29.93 -92.51%). Although the occurrence of H. canis infection was low, significative hematological alterations were observed in most infected dogs. Coinfection with Babesia spp. and E. canis was detected in two dogs and the hematological alterations cannot be attributed exclusively to H. canis in these animals. Longitudinal studies would be of fundamental importance to determine the causality of these alterations. These results highlight the importance of differential diagnosis in dogs when there is clinical suspicion of infection by hemoparasites, since the hematological changes in dogs infected by H. canis are quite variable.

Blood samples were collected from 99 domestic dogs from the urban and rural areas of the Lábrea municipality, state of Amazonas, Brazil. Canine serum samples were tested by immunofluorescence assay against Rickettsia spp., which revealed that only 3.0% (1/33) and 7.6% (5/66) of the dogs from urban and rural areas, respectively, reacted positively to at least one Rickettsia species. DNA was extracted from canine blood and tested by a battery of PCR assays targeting protozoa of the genera Babesia and Hepatozoon, and bacteria of the genera Rickettsia and Ehrlichia and family Anaplasmataceae. All samples were negative in the PCR assays targeting the genera Babesia, Hepatozoon, Ehrlichia and Rickettsia. For Anaplasmataceae, 3% (1/33) and 39.4% (26/66) of the urban and rural dogs, respectively, yielded amplicons that generated DNA sequences 100% identical to the corresponding sequence of Wolbachia endosymbiont of Dirofilaria immitis. Because of these results, all canine DNA samples were further tested in a PCR assay targeting filarial nematodes, which was positive for 18.2% (6/33) and 57.6% (38/66) urban and rural dogs, respectively. Filarial-PCR products generated DNA sequences 100% identical to D. immitis. While tick-borne infections were rare in Lábrea, D. immitis infection rates were among the highest reported in South America.Keywords: Dogs, ticks, Rickettsia, Wolbachia, Dirofilaria immitis, Amazon. ResumoAmostras de sangue foram coletadas de 99 cães domésticos de áreas urbana e rural do município de Lábrea, estado do Amazonas. Soros caninos foram testados pela técnica de imunofluorescência indireta contra Rickettsia spp., resultando em apenas 3,0% (1/33) e 7,6% (5/66) de cães soropositivos nas áreas urbana e rural, respectivamente. DNA foi extraído do sangue canino e testado por diferentes protocolos da PCR para detecção de protozoários dos gêneros Babesia e Hepatozoon, e bactérias dos gêneros Rickettsia e Ehrlichia e da família Anaplasmataceae. Todas as amostras foram negativas nos protocolos de PCR para os gêneros Babesia, Hepatozoon, Ehrlichia e Rickettsia. Para Anaplasmataceae, 3% (1/33) e 39,4% (26/66) dos cães de áreas urbana e rural, respectivamente, geraram sequências de DNA 100% idênticas ao endosimbionte Wolbachia de Dirofilaria immitis. Posteriormente, as amostras foram testadas pela PCR para nematódeos filarídeos, resultando em 18,2% (6/33) e 57,6% (38/66) de amostras positivas nas áreas urbana e rural, respectivamente. Os produtos geraram sequências de DNA 100% idênticas a D. immitis. Em contraste com várias outras regiões do Brasil, infecções transmitidas por carrapatos foram raras em Lábrea. Por outro lado, as frequências de infecção por D. immitis estiveram entre as mais altas relatadas na América do Sul.

Özet: Bu canis vogeli ve B. gibsoni izolatlarının 18S rRNA gen bölgesine ait sekanslarının dünyadaki aynı türden diğer izolatlarla ikili kıyaslamalarında %1,4±0,2, %0,3±0,2 ve %0,9±0,3 genetik farklılık saptanmıştır. E. canis için 16S rRNA gen bölgesi yönünden üç izolatın kendi aralarında %100 identik olduğu, dünyadaki izolatlarla ise %0,1 farklılık gösterdiği; A. phagocytophilum için ankA gen bölgesi yönünden üç izolatın kendi aralarında %99,8±0,2 identik olduğu, dünyadaki izolatlarla ise %0,9±0,3 farklılık gösterdiği; H. canis için 18S rRNA geni yönünden iki izolatın kendi aralarında %100 identik olduğu, dünyadaki izolatlarla ise %0,2±0,1 farklılık gösterdikleri tespit edilmiştir. Sonuç olarak bu çalışmayla Kayseri yöresinde köpeklerde kene kaynaklı protozoon ve rickettsial enfeksiyonların moleküler prevalansları saptanmış ve enfeksiyonlara yol açan türlerin çeşitli gen bölgeleri analiz edilerek moleküler karakterizasyonları yapılmıştır. Tüm izolatların Genbank kayıtları gerçekleştirilmiştir.Anahtar sözcükler: Anaplasmosis, babesiosis, ehrlichiosis, hepatozoonosis, köpek, moleküler karakterizasyon, Real Time PCR. B. gibsoni, A. phagocytophilum, H. canis, and B. canis vogeli was detected as 14.5%, 12.0%, 9.0%, 7.8%, 5.3%, and 2.3%, respectively while B.canis rossi was not detected in the examined samples. 182 (89.7%) out of the 400 samples were found to be infected with a single parasite species, and 21 (10.3%) were found to be infected with two species. According to the pairwise comparisons of 18S rRNA gene region of the isolates under B. canis canis, B.canis vogeli and B. gibsoni, 1.4±0.2%, 0.3±0.2%, and 0.9±0.3% genetic distance were detected with the other similar isolates from the world, respectively. Based on the 16S rRNA gene sequence alignments, 100% identity was found among the 3 isolates of E. canis while 0.1% genetic difference was determined with the isolates from the world. With respect to ankA gene region of A. phagocytophilum, 99.8±0.2% identity and 0.9±0.3% genetic difference were found among the 3 isolates obtained from Kayseri region and the isolates from the world, respectively. The 2 H. canis isolates were showed 100% identity to each other and 0.2±0.1% genetic difference were determined with the other isolates from the world with respect to 18S rRNA gene region. In conclusion, the molecular prevalence of tick-borne protozoon and rickettsial infections in dogs in Kayseri region was determined and the molecular characterizations of the obtained isolates were performed by analyzing the various gene regions in this study. All isolates were recorded to GenBank.
