Protective effects of Tadalafil and darbepoetin against ischemia - reperfusion injury in a rat testicular torsion model

Abstract: Objectives:To evaluate protective effects of darbepoetin and tadalafil against ischemia-reperfusion injury in ipsilateral and contralateral testicle.Materials and Methods:Thirty 3-month-old adult male Wistar-Albino rats were randomly divided into 5 groups (A-E). Sham operation was performed in the first group. In Group B, rats did not received any medication after creating 720 degrees torsion of the left testis. The rats in Group C, D and E received darbepoetin, tadalafil, and darbepoetin/tadalafil combination… Show more

“…Testicular damage in the torsion/detorsion groups was evident compared to the other groups. It is possible to see similar evidence in the previous study (Yildirim et al, 2018). The healing effect of chrysin on histopathological injury has been reported in the cerebral ischaemia/reperfusion study (Durak et al., 2016).…”

“…It was determined that expression level of caspase‐3 and caspase‐8 was the most severe in T‐2h/D‐2h group, moderate in T‐2h/D‐24h group, mild in T‐2h/D‐2h + CR group and very mild in T‐2h/D‐24h + CR group. In particular, our result in T‐2h/D‐2h group supports the 25‐min renal ischaemia study result (caspase‐3 and caspase‐8 expression) and the testicular ischaemia study result (caspase‐3 expression) (Sekerci et al, 2017; Yildirim et al., 2018; Zhang et al., 2006). Chrysin application reduced both caspase‐3 and caspase‐8 expression levels in our study.…”

“…Germinal cell death may follow testicular ischaemia due to the accumulation of toxic metabolites leading to damage in DNA, proteins and lipid contents of testicular cells with an increase in the production of vascular cell adhesion molecule‐1 (VCAM‐1), which enhances leucocyte proliferation and migration with the synthesis of different pro‐inflammatory cytokines, as tumour necrosis factor‐α (TNF‐α), causing germ cell apoptosis and testicular atrophy (Sperling, Krege, Lümmen, & Rübben, ; Vaos & Zavras, ; Yildirim et al, ).…”

“…720‐degree rotation), as it causes irreversible ischaemic damage and gonad loss in the testis (Asgari et al, 2006). Despite successful surgical intervention, 40%–60% of testicular atrophy may develop into infertility in the following years (Yildirim et al, 2018).…”

“…Nevertheless, except for cooling the scrotum, no treatment has been successfully performed in clinical practice to date (Visser and Heyns, 2003). In experimental studies, many substances, such as sildenafil, nifedipine, ozone, darbepoetin and tadalafil, have been wasted to prevent the damage due to I/R and detorsion which may result in infertility (Yildirim et al, 2018; Mestrovic et al, 2014; Beheshtian et al, 2008; Tusat et al, 2017).…”
