The hOGG1 Ser326Cys gene polymorphism and susceptibility for bladder cancer: a meta-analysis

Abstract: Objective:To assess the susceptibility of the hOGG1 genetic polymorphism for bladder cancer and evaluate the impact of smoking exposure.Materials and Methods:Articles included in PubMed, Medline and Springer databases were retrieved using the following key words: “human 8-oxoguanine DNA glycosylase”, “OGG”, “OGG1”, “hOGG1”, “genetic variation”, “polymorphism” , “bladder cancer”, and “bladder carcinoma” to Meta-analysis was performed to detect whether there were differences between the bladder cancer group and … Show more

“…Significant results were observed in lung ( 59 , 68 ), head and neck ( 69 , 70 ), colorectal ( 51 , 71 ) and gallbladder cancer ( 65 , 72 ). However, several previous meta-analyses ( 68 , 70 , 73 76 ) did not reveal any significant association between this OGG1 polymorphism and other cancers risk.…”

“…These workers had the higher levels of urinary 1-OHP as compared to reference values, indicating high exposure to PAH. Urinary 1-OHP levels in occupational workers were higher than those of control population as suggested by Wenjuan et al [ 19 ] . These observations are also in accordance with those reported previously by Fu et al [ 20 ] and Nguyen et al [ 21 ] who described the toxicity of PAH as a consequence of imbalance between pro-oxidant and antioxidant homeostasis, the so called oxidative stress, which ultimately leads to oxidative damage.…”

“…126 Although having a clear role in mutagenic processes, OGG1 -null mice showed only moderate increases in malignancy rate, likely due to effective alternative damage repair pathways. 127 Evidence from multiple meta-analyses 128-130 of OGG1 involvement in UBC cancerogenesis is contradictory, and, when having a genuine effect, is more likely to play a supporting role in a multi-stage process rather than being the main cause of it. 127 It is also probable that the establishment of the type and direction of genetic associations requires larger populations (underscoring sufficient sample sizes for different ethnicities), not yet available to researchers.…”
