The aim of this study was to microscopically re-evaluate the melanocytic lesions diagnoses established by the Animal Pathology Laboratory of the Federal University of Uberlândia, in the state of Minas Gerais, Brazil, over a period of eleven years; in addition, to perform a comparative analysis between the conventional histopathological (CH) method and the use of the melanin bleaching (MB) technique with potassium permanganate, sulfuric acid, and oxalic acid solutions. The results of the MB method presented a disagreement in 24.32% of the diagnosis previously by CH, with low agreement (61.0%) and low Kappa coefficient (0.2267). Melanoma was the most frequent lesion, more frequent in elderly and non-breed female dogs. The most frequent melanoma location was in the cutaneous tissue. The presence or absence of a pagetoid spread in cutaneous samples, distribution of melanin, pattern of cell layout, cell morphology, degree of cellular atypia, and the number of mitoses verified after MB were the most important criteria to confirm the diagnosis of malignancy or benignity of the lesions. Evaluating pathologists considered MB to be essential for the majority of diagnoses and an efficient complementary method for the diagnosis of melanocytic lesions, even in cases with a moderate degree of pigmentation.

The aim of this study was to verify the applicability and accuracy of B-mode ultrasonography in detecting malignancy in dog cutaneous neoplasms. Forty-two neoplasms (12 benign and 30 malignant) of mesenchymal, round cells, epithelial and melanocytic origins from 24 dogs of different breeds and ages were included. The ultrasound evaluation was performed with a linear multi-frequency transducer (7.0 to 12MHz), with frequency dependent on the mass dimension. Ultrasonographic characteristics of echogenicity (hypo/hyperechogenic), echotexture (homogeneous/heterogeneous), regularity, invasiveness in adjacent tissues were classified. Dimensions were also measured to calculate the depth/width ratio. Neoplasms were classified as malignant or benign after cytological and/or histopathological analysis and the results were associated with ultrasound characteristics. There was a significant association (P<0.05) between malignancy and echogenicity, echotexture and invasiveness in adjacent tissues, so that 84.6% of hypoechogenic neoplasms, 76.9% of heterogeneous masses and 88.2% of invasive neoplasms were classified as malignant. However, for all these associations, moderate predictive values were obtained, which may be due to the small experimental number included in this study. Therefore, although it has been observed that hypoechogenic, heterogeneous and invasive neoplasms were more prone to malignancy, these findings should be used with caution until new studies are developed with a greater number and variety of cutaneous neoplasms in dogs.

Mutational status of naevus-associated melanomas

Introduction Melanoma origin has always been a debated subject, as well as the role of adjacent melanocytic nevi. Epidemiological and histopathological studies point to melanomas arising either de novo or from a nevus. Methods Sixty-one melanomas found in association with a preexisting nevus were microdissected, after careful selection of cell subpopulations and submitted to Sanger sequencing of the BRAF, NRAS, C-KIT, PPP6C, STK19 and RAC1 genes. Each gene was evaluated twice in all samples by sequencing or by sequencing and another confirmation method, allele-specific fluorescent polymerase chain reaction (PCR) and capillary electrophoresis detection, or by SNaPshot Analysis. Only mutations confirmed via two different molecular methods or twice by sequencing were considered positive. Results The majority of cases presented concordance of mutational status between melanoma and the associated nevus for all 6 genes (40/60; 66.7%). Nine cases presented concomitant BRAF and NRAS mutations, including one case, in which both the melanoma and the adjacent nevus harbored V600E and Q61K double mutations. In two cases, both melanoma and associated nevus, located on acral sites were BRAF mutated, including an acral lentiginous melanoma. Conclusions This is the largest nevus-associated melanoma series molecularly evaluated to our knowledge. The majority of melanomas and adjacent nevi in our sample share the same mutational profile, corroborating the theory that the adjacent nevus and melanoma are clonally related and that melanoma originated within a nevus.
